摘要
甜菜(Beta)M14含有野生白花甜菜的染色体,这使得花期甜菜M14的蛋白质表达存有差异。本研究提取栽培甜菜和甜菜M14开花后1 h的花蕾的蛋白质,利用双向电泳分离蛋白质,使用凝胶分析软件分析电泳图谱,观察到甜菜M14中有5个蛋白点存在表达差异,甜菜M14中1个蛋白点缺失,另有少量蛋白质存在表达丰度的增强或减弱。选取其中2个差异蛋白点进行质谱检测,并在NCBI数据库里进行分析,结果表明其中一个蛋白差异点与核酮糖1,5-二磷酸羧化酶或其小亚基有极高的同源性和相似性。本研究为进一步解释甜菜M14染色体数目的增加影响其蛋白质表达水平提供了新的实验证据。
Sugar beet(Beta)M14 contains chromosomes of Beta colloroflora,which makes the protein expression of beet M14 different at flowering stage.We extracted cultivated beet and sugar beet M141 h after flowering buds of protein,using two-dimensional electrophoresis separation of proteins,and used gel electrophoresis analysis software to analyze the electrophoretic pattern.It was observed that there were five protein spots differentially expressed in sugar beet M14,one protein spot was absent in sugar beet M14,and a few protein expressions were enhanced or decreased in abundance.Two differential protein spots were selected for mass spectrometry detection,results were analyzed in the NCBI database.One protein spot has a very high similarity and homology with ribulose 1,5-bisphosphate carboxylase or its small subunit.This study provides new experimental evidence for further explaining the variation of chromosome number in the progeny of M14 on the difference of protein expression.
