摘要
目的:观察松郁安神方对失眠大鼠睡眠时相的影响。方法:采用腹腔注射对氯苯丙氨酸(Para-chlorophenylalanine,PCPA)建立失眠大鼠模型,用松郁安神方进行干预,通过动物睡眠生物解析系统,记录脑电(Electroencephalogram,EEG)和肌电(Electromyogram,EMG),分析睡眠觉醒时间、非快速眼动睡眠(Non-rapid Eyemovement Sleep,NREM)时间和快速眼动睡眠(Rapid Eyemovement Sleep,REM)时间,观察大鼠睡眠时相变化。结果:PCPA可明显减少大鼠白天NREM、REM睡眠时间,增加觉醒时间,给予松郁安神方后,失眠大鼠白天时段NREM、REM睡眠时间增加,觉醒时间减少,与PCPA模型组比较,差异有统计学意义(P<0.05或P<0.01)。结论:松郁安神方能改善PCPA致失眠大鼠的睡眠,其主要通过增加白天的NREM和REM睡眠时间改善睡眠。
Objective:To observe the effect of Songyu Anshen formula on sleep phase in insomnia rats.Methods:Para-chlorophenylalanine(PCPA)was injected intraperitoneally to establish the model of insomnia rats.The rats were administrated intragastrically with Songyu Anshen formula.The changes of sleep phases in rats were observed by recording electroencephalogram(EEG)and electromyogram(EMG),and the amount of waking,non-rapid eyemovement(NREM)sleep and rapid eyemovement(REM)sleep were analyzed.Results:PCPA can significantly reduce NREM and REM sleep duration,and increase the wakefulness time in daytime.Songyu Anshen formula can significantly increase NREM and REM sleep duration,and reduce the wakefulness time of insomnia rats induced by PCPA during daytime(P<0.05,P<0.01,P<0.001).Conclusion:Songyu Anshen formula can improve sleep quality of insomnia rats,mainly by increasing NREM and REM sleep duration in the daytime.
