奶牛养殖场榨乳环节不同取样点细菌菌群的16S rRNA分子生物学分析 被引量:1

摘要 榨乳环节是奶牛养殖的关键环节之一,养殖场原奶质量直接影响消费者的健康,为了解奶牛养殖场榨乳环节不同取样点的微生物多样性与差异性,试验选择河北省唐山地区某规模化奶牛养殖场的6个典型榨乳环节,即牛乳头消毒前(C1)、牛乳头消毒后(C2)、采奶罩杯(E)、储奶设备(E2)、储奶罐奶(M1)、运输车奶(M2)为取样点,采用高通量测序技术研究了各取样点细菌的多样性特征和丰富度差异。结果表明:从6个不同取样点共获得1 969 296条原始序列,有效序列1 909 879条,聚成37 510个OTUs,注释到33门80纲129目226科457属水平物种,各取样点优势菌属有不动杆菌属、节细菌属、鞘氨醇杆菌属、球菌属、棒状杆菌属;C1取样点的细菌群落丰富度最高,M1取样点的细菌群落均匀度最好。说明奶牛养殖场榨乳环节各取样点菌群多样性存在差异,且丰富度与均匀度明显不同。 Milk collection is one of the key links in dairy farming. Farm raw milk quality directly affect the health of consumers. In order to understand the diversity and differences of microorganisms at different sampling sites during milking process in dairy farms, 6 typical milking sites(including cattle nipple before disinfection(C1), nipple after disinfection(C2), milking cup(E), tube of milk storing cans(E2) and the different links of raw milk samples, milk of storage tank(M1) and milk in the transporters(M2)) were selected from a large-scale dairy farm in Tangshan, Hebei Province. The diversity and richness of bacteria in these sampling sites were studied by high-throughput sequencing technology. The results showed that a total of 1 969 296 sequences were obtained from 6 different sampling sites, among which 1 909 879 effective sequences were clustered into 37 510 OTUs, which were annotated to 33 phyla, 80 classes, 129 orders, 226 families, 457 genera. Dominant bacteria genera at different sampling sites include Acinetobacter, Arthrobacter, Sphingobacterium, Macrococcus and Corynebacterium. The richness of the bacterial community was the highest at C1 sample point, The bacterial community uniformity was best at the M1 sample point. The results indicated that there were differences in the diversity, richness and evenness of various sample bacteria in the milking process of dairy cattle farms.
