摘要
目的探讨Toll样受体4(TLR4)和TLR9在卵巢癌组织中的表达及其临床意义,为卵巢癌的临床诊疗及作用机制阐释提供参考依据。方法选取卵巢癌手术患者60例、因子宫或者卵巢良性病变而行手术治疗的患者30例,采用免疫组化(SP)法检测卵巢癌病灶及癌旁组织、正常卵巢组织中TLR4、TLR9蛋白的表达,采用逆转录聚合酶链反应(RT-PCR)检测TLR4 mRNA、TLR9 mRNA水平,分析TLR4、TLR9蛋白表达与卵巢癌患者临床病理特征的关系;运用秩相关分析探讨卵巢癌组织中TLR4与TLR9蛋白的关系。结果卵巢癌病灶组织中TLR4蛋白表达的阳性率为61.67%,TLR9蛋白表达的阳性率为93.33%,TLR4、TLR9蛋白阳性表达及TLR4 mRNA、TLR9 mRNA在卵巢癌组织、癌旁组织、正常卵巢组织中的表达均依次降低,差异均有统计学意义(P<0.01)。不同肿瘤分化程度、FIGO分期、淋巴结转移是影响卵巢癌组织中TLR4蛋白表达的主要因素,差异均有统计学意义(P<0.05);不同肿瘤分化程度、淋巴结转移是影响TLR9蛋白表达的主要因素,差异均有统计学意义(P<0.05)。卵巢癌组织中TLR4与TLR9两种蛋白表达具有正相关性(P<0.01)。结论TLR4和TLR9在卵巢癌组织中高表达,TLR4、TLR9表达与肿瘤分化程度、淋巴结转移情况有关,且两者呈正相关性。提示TLR4、TLR9可能参与了卵巢癌的发生、发展过程,这也为卵巢癌的免疫治疗提供了依据和支撑。
Objective To explore the expressions and clinical significance of Toll-like receptor 4(TLR4)and TLR9 in ovarian cancer,provide a reference basis for explanation of clinical diagnosis,treatment,mechanism of ovarian cancer.Methods Sixty patients treated by ovarian cancer surgery and thirty patients treated by surgery due to uterine or ovarian benign diseases were selected,immunohistochemical SP method was used to detect the expressions of TLR4 protein and TLR9 protein in ovarian cancer lesion,adjacent tissue of ovarian cancer,and normal ovarian tissue.RT-PCR was used to detect the levels of TLR4 mRNA and TLR9 mRNA.The relationships between expressions of TLR4 protein and TLR9 protein and clinicopathological characteristics of ovarian cancer patients were analyzed.Rank correlation analysis was used to explore the correlation between TLR4 protein and TLR9 protein in ovarian cancer patients.Results The positive expression rates of TLR4 protein and TLR9 protein in ovarian cancer lesion were 61.67%and 93.33%,respectively.The positive expression rates of TLR4 protein and TLR9 protein,the expression levels of TLR4 mRNA and TLR9 mRNA in ovarian cancer lesion,adjacent tissue of ovarian cancer,and normal ovarian tissue decreased gradually,there were statistically significant differences(P<0.01).Differentiation degree,FIGO stage,and lymph node metastasis were main influencing factors of TLR4 protein expression in ovarian cancer(P<0.05).Differentiation degree and lymph node metastasis were main influencing factors of TLR9 protein expression in ovarian cancer(P<0.05).In ovarian cancer tissue,TLR4 protein expression was positively correlated with TLR9 protein expression(P<0.01).Conclusion TLR4 and TLR9 are highly expressed in ovarian cancer tissue.The expressions of TLR4 and TLR9 are correlated with tumor differentiation degree and lymph node metastasis,there is positive correlation between TLR4 and TLR9,which indicates that TLR4 and TLR9 may be involved in occurrence and development of ovarian cancer,this study provides evidenc
