摘要
目的观察马齿苋多糖对反流性食管炎(reflux esophagitis,RE)模型大鼠胃排空功能和幽门括约肌的收缩功能,以及血清胃肠激素和食管组织匀浆细胞因子的影响,并初步探讨其可能机制。方法采用刺激法+导尿管球囊扩张法建立反流性食管炎动物模型,以附子理中丸为阳性对照药。给予大鼠营养性半固体糊,以胃排空率为指标,分别观察低剂量(7.5 g/kg)、中剂量(15 g/kg)和高剂量(30 g/kg)马齿苋多糖对大鼠胃排空率的影响。通过记录幽门括约肌自发收缩曲线,观察3种不同浓度马齿苋多糖对幽门括约肌收缩功能的影响。用酶联免疫吸附法分别测定大鼠血清血管活性肠肽(VIP)、Ghrelin、胃泌素(GAS)等胃肠激素和胃动素(MTL)以及食管组织白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、干扰素因子-γ(IFN-γ)等细胞因子含量。结果3个剂量组马齿苋多糖呈剂量依赖性地增强了大鼠胃排空率(P<0.05);3种不同浓度马齿苋多糖呈剂量依赖性地降低了幽门括约肌收缩曲线的平均振幅(P<0.05);与模型组比较,马齿苋多糖高剂量组血清Ghrelin、GAS、MTL水平升高(P<0.05),VIP水平下降(P<0.05);食管组织匀浆IL-1β、IL-6、TNF-α、IFN-γ等细胞因子含量下降(P<0.01)。结论马齿苋多糖可改善RE模型大鼠的胃排空功能,抑制幽门括约肌收缩,改善胃肠动力,减轻胃反流物对食道黏膜刺激;此作用可能与升高血清Ghrelin、GAS和MTL水平,降低VIP水平,降低食管组织匀浆IL-1β、IL-6、TNF-α、IFN-γ含量有关。
Objective To observe the effects of Portulaca oleracea polysaccharide(POP)on gastric emptying function,pylorus sphincter contractile function,gastrointestinal hormones and inflammatory factors in reflux esophagitis model rats,and to preliminary explore its possible mechanism.Methods The animal model of reflux esophagitis was established by stimulation method and catheter balloon dilatation method,with Fuzi Lizhong Pill as a positive control drug.Rats were given a nutritious semi-solid paste,and the gastric emptying rate was used as an indicator,the effects of low dosage(7.5 g/kg),middle dosage(15 g/kg)and high dosage(30 g/kg)POP on gastric emptying rate in rats were observed.By recording the contraction curve of the pylorus sphincter,the effects of three different concentrations of POP on the contraction of the lower esophageal sphincter were determined.Enzyme-linked immunosorbent assay was used to measure rat gastrointestinal hormones such as:serum vasoactive intestinal peptide(VIP),Ghrelin,gastrin(GAS)and motilin(MTL)and esophageal tissue interleukin-1β(IL-1β),interleukin-6(IL-6),tumor necrosis factor-α(TNF-α)and interferon factor-γ(IFN-γ)were measured.Results POP in three dose groups enhanced the gastric emptying rate of rats dose-dependently(P<0.05);Three concentrations of it increased the average amplitude of the contraction curve of the pylorus sphincter dose-dependently(P<0.05);Compared with the model group,serum Ghrelin,gastrin,and motilin levels in the high-dose POP group increased(P<0.05),and vasoactive intestinal peptide levels decreased(P<0.05);The contents of IL-1β,IL-6,TNF-αand IFN-γin esophageal homogenate were decreased(P<0.01).Conclusion POP can improve the gastric emptying function of reflux esophagitis model rats,promote the contraction of the pylorus sphincter,improve gastrointestinal motility,and reduce the stimulation of gastric reflux to the esophageal mucosa.These effects may be related to increased serum Ghrelin,gastrin and motilin levels and decreased VIP,decreased esophageal ho
