摘要
目的建立防风饮片标准汤剂的HPLC指纹图谱,结合化学计量学方法对其进行质量研究。方法制备15批次防风饮片标准汤剂,建立其HPLC指纹图谱,采用相似度评价结合聚类分析(cluster analysis,CA)、主成分分析(principal component analysis,PCA)、偏最小二乘判别分析(partial least squares discriminant analysis,PLS-DA)进行系统评价,同时测定了防风中4种主要成分的含量变化。结果建立了防风标准汤剂HPLC指纹图谱,其相似度>0.92,确定了8个共有峰。CA的结果表明不同产地的防风质量概貌存在差异,同一产地药材质量较为稳定。PLS-DA筛选出4个差异标志物(VIP>1)分别为3号,6号峰,4号峰,5号峰。15批次防风饮片标准汤剂共有模式下的4种主要成分分别为升麻素苷,升麻素,5-O-甲基维斯阿米醇苷,亥茅酚苷,其转移率依次为52.53%-88.03%,48.83%-82.85%,41.90%-80.45%,37.69%-84.49%。15批次防风饮片标准汤剂干膏得率36.09%-49.45%,pH值5.01-5.75。结论本研究建立的质量评价方法适用于防风饮片标准汤剂的质量控制,可为防风配方颗粒及相关经典名方的研究提供参考。
Objective To establish HPLC fingerprints of standard decoctions of Radix Saposhnikoviae decoction pieces,and to study their quality by chemometric methods.Methods A total of 15 batches of standard decoctions of Radix Saposhnikoviae decoction pieces were prepared and their HPLC fingerprints were established.Similarity evaluation combined with cluster analysis(CA),principal component analysis(PCA)and partial least squares discriminant analysis(PLS-DA)was applied to perform comprehensively evaluation.Content changes of 4 major components in Radix Saposhnikoviae were measured at the same time.Results The HPLC fingerprints of the standard decoctions of Radix Saposhnikoviae were established,whose similarity was higher than 0.92,and 8 common peaks were determined.Results of the cluster analysis showed that there were differences in quality general pictures of Radix Saposhnikoviae from different producing areas,but the quality was relatively stable in medicinal materials from the same origin.Four differential markers(VIP>1)were screened out by PLS-DA,which were peak 3,peak 6,peak 4 and peak 5,respectively.Under common patterns,there were 4 major components in the 15 batches of standard decoctions of Radix Saposhnikoviae decoction pieces,including Prim-O-glucosylcimifugin,cimifugin,5-O-methylvisamminol and Sec-O-glucosylhamaudol,and their transfer rates were 52.53%-88.03%,48.83%-82.85%,41.90%-80.45%and 37.69%-84.49%.The dry extract yielding rate of the 15 batches of standard decoctions of Radix Saposhnikoviae decoction pieces was 36.09%-49.45%,and the pH was 5.01-5.75.Conclusion This study established a quality evaluation method that can be applied to quality control of standard decoctions of Radix Saposhnikoviae decoction pieces.It can provide reference for study on formula granules of Radix Saposhnikoviae and relevant classical formulas.
