60Co-γ射线辐射灭菌对连翘药材指纹图谱及有效成分的影响 被引量:5

摘要 目的:探索60Co-γ射线辐照灭菌对连翘药材有效成分的影响及辐照前后指纹图谱的变化,为连翘辐照灭菌提供指导依据。方法:采用Agilent ZORBAX SB-C18色谱柱(250 mm×4.6 mm,5μm),以乙腈-0.1%甲酸溶液为流动相,梯度洗脱;流速:1.0 ml·min^(-1);检测波长:280 nm;柱温:30℃,建立连翘药材HPLC指纹图谱。采用SPSS 22.0软件比较3,6,10 k Gy剂量下60Co-γ射线辐照对连翘中芦丁、连翘酯苷A、连翘酯苷B、连翘苷含量,及对指纹图谱相似度的影响。结果:芦丁、连翘酯苷B、连翘酯苷A、连翘苷的进样量分别在0.046~2.297,0.030~1.465,0.294~14.553,0.091~4.504μg范围内线性关系良好(r≥0.999 1);平均回收率分别为99.24%(RSD=0.59%),100.49%(RSD=0.49%),97.61%(RSD=0.74%),102.25%(RSD=0.33%)(n=9)。当辐照剂量不超过6 kGy时,连翘药材中的芦丁、连翘酯苷A、连翘酯苷B、连翘苷含量变化差异无统计学意义(P>0.05)。结论:60Co-γ射线辐照灭菌对连翘药材有效成分和指纹图谱无影响,可作为其有效灭菌手段。 Objective:To investigate the influence of60Co-γirradiation sterilization on the contents of effective components and the change of fingerprint before and after radiation sterilization,so as to provide guide for the sterilization of Forsythiae fructus.Methods:The analysis was performed on an Agilent ZORBAX SB-C18(250 mm×4.6 mm,5μm) column with gradient elution of acetonitrile and0.1%formic acid solution at the flow rate of 1.0 ml·min^(-1).The detection wavelength was set at 280 nm,and the column temperature was 30℃.The fingerprint was established.The content changes of rutin,forsythoside A,forsythoside B and forsythin in Forsythiae fructus before and after60Co-γirradiation sterilization at dose of 3,6 and 10 k Gy were compared and analyzed by SPSS 22.0 software,and the similarities of fingerprint were also compared.Results:The injection volume of rutin,forsythoside B,forsythoside A and forsythin had a good linear relationship in the range of 0.046-2.297,0.030-1.465,0.294-14.553 and 0.091-4.504μg,respectively (r≥0.999 1).The average recovery rates were 99.24%(RSD=0.59%),100.49%(RSD=0.49%),97.61%(RSD=0.74%),102.25%(RSD=0.33%)(n=9).When the radiation dose was less than 6 k Gy,the contents and fingerprint of rutin,forsythoside A,forsythoside B and forsythin had no significant changes(P>0.05).Conclusion:60Co-γray radiation sterilization has no notable effect on the contents and fingerprint of Forsythiae fructus.and it can be used for the sterilization of Forsythiae fructus.
