基于系统药理学方法研究川楝素对肝癌的抗肿瘤机制 被引量:1

摘要 目的基于系统药理学方法,系统研究川楝素对肝癌的抗肿瘤机制。方法应用STITCH、SEA和TargetNet数据库预测川楝素的可靠靶点,应用DisGeNET数据库获得关于肝肿瘤的作用靶点,通过HINT和STRING数据库获得蛋白-蛋白互作信息,通过DAVID数据库获得GO功能和KEGG富集分析结果,通过Cytoscape软件可视化生物网络。结果川楝素对肝癌的抗肿瘤机制中比较重要的通路有:癌症通路、甲状腺激素信号通路、PI3K-Akt信号通路、前列腺癌和癌症中蛋白聚糖等;比较重要的靶点有:AKT1、PIK3R1、CTNNB1、GRB2和PIK3R2等。结论川楝素对肝癌的抗肿瘤机制具有多靶点和多通路的性质,本研究从整体上为川楝素对肝癌的抗肿瘤机制研究提供了参考。 Objective To systematically study the antitumor mechanism of toosendanin on hepatocellular carcinoma(HCC)based on systematic pharmacology.Methods The reliable targets of toosendanin were predicted by using the databases of STITCH,SEA and TargetNet.The targets of liver tumor were obtained by using the database of DisGeNET.The protein-protein interaction information was obtained by using the databases of HINT and STRING.The GO function and KEGG enrichment analysis results were obtained by using the database of DAVID.The biological network was visualized by using the software of Cytoscape.Results In the anti-tumor mechanism of toosendanin on liver cancer,the more important pathways are:cancer pathway,thyroid hormone signaling pathway,PI3K Akt signaling pathway,prostate cancer and proteoglycan in cancer;the more important targets are:AKT1,PIK3R1,CTNNB1,Grb2 and pik3r2.Conclusion The anti-tumor mechanism of toosendanin on liver cancer has the characteristics of multi-target and multi-channel.This study provides a reference for the study of the anti-tumor mechanism of toosendanin on liver cancer as a whole.
