摘要
为了解多重耐药基因cfr、optrA在猪源葡萄球菌中的流行分布,对来自广东省河源市和上海市3个猪场的280份鼻腔拭子样品,使用氟苯尼考(10μg/mL)甘露醇平板进行细菌分离,PCR检测cfr和optrA耐药基因,并通过gap基因进行菌种鉴定,采用琼脂稀释法对16种常用抗菌药物进行药敏试验。结果显示,本研究共分离获得38株葡萄球菌(分离率为13.57%),包括25株cfr阳性菌株(检出率为8.93%)和15株optrA阳性菌株(检出率为5.36%),其中2株葡萄球菌同时检出cfr及optrA基因。gap基因鉴定表明,15株optrA阳性葡萄球菌均为松鼠葡萄球菌,23株cfr阳性葡萄球菌成功分为4个不同葡萄球菌亚种,包括12株松鼠葡萄球菌、8株腐生葡萄球菌、2株马胃葡萄球菌及1株科氏葡萄球菌。药敏结果显示,38株cfr/optrA葡萄球菌均为多重耐药菌株,均对氟苯尼考、克林霉素和泰妙菌素耐药,且对部分药物表现出较高耐药率,如沃尼妙林(94.7%)、氨苄西林(92.0%)及四环素(89.5%),但所有菌株对万古霉素敏感。对比在不同生长阶段分离葡萄球菌中cfr及optrA的检出率,cfr和optrA基因主要流行于育肥猪。结果表明,3个猪场葡萄球菌cfr和optrA的检出率高,耐药情况严重,应加强抗菌药物在养殖过程中规范使用以减缓耐药性的产生和传播。
To investigate the prevalence and distribution of resistance genes cfr and optrA in Staphylococci from three swine farms in Heyuan city of Guangdong province and Shanghai city.A total of280 nasal swab samples were collected and the Staphylococcus isolates was isolated using mannitol agar containing 10 μg/mL florfenicol.All isolates were screened for the presence of cfr and optrA resistance genes by PCR amplification and staphylococcal species were identified by amplifying gap gene.Antimicrobial susceptibility test was determined using agar dilution method for all cfr or optrA-positive Staphylococci.The results showed that a total of 38 porcine Staphylococcus isolates were obtained.PCR amplification indicated that 25(8.93%) and 15(5.36%) isolates were positive for cfr and optrA genes,respectively.Two isolates were positive for cfr and optrA genes.It is noteworthy that all optrA-positive isolates were identified as Staphylococcus sciuri.The 23 cfr-positive isolates were successfully classified as four Staphylococcus spp.including 12 Staphylococci sciuri,8 Staphylococci saprophyticus,2 Staphylococci equorum and 1 Staphylococci cohnii.The antimicrobial susceptibility results showed that all cfr or optrA-positive Staphylococcus isolates were multidrug-resistant bacteria,all Staphylococcus isolates were resistant to florfenicol,clindamycin and tiamulin,and the drug resistance rate to valnemulin,ampicillin,tetracycline were 94.7%,92.0% and 89.5%,respectively.All isolates were sensitive to vancomycin.Compared with the detection rate of cfr and optrA genes among Staphylococcus isolates at different growth stages,the cfr and optrA genes were mainly prevalent in Staphylococci from fattening pigs.The experimental results indicated that the antimicrobial resistance of porcine Staphylococci was serious and the prevalence of cfr or optrA was high in three swine farms.It is necessary to strengthen the criterion use of antibiotics in livestock and poultry breeding to reduce the generation and spread of antimicrobial resistance b
