摘要
目的:通过网络药理学方法探讨黄芪桂枝五物汤治疗类风湿关节炎的分子作用机制。方法:利用TCMSP数据库检索、筛选出黄芪桂枝五物汤中药活性成分和作用靶点,使用GeneCards预测类风湿关节炎疾病靶点,构建中药活性成分-靶点网络图,将获得的靶点导入STRING数据库构建蛋白-蛋白相互作用网络,对靶点基因进行GO和KEGG富集分析。结果:共筛选出黄芪桂枝五物汤中药活性成分74种,作用于217个靶点,类风湿关节炎疾病靶点4329个,从而获得药物和疾病的共同靶点146个。GO分析涉及对脂多糖的反应、对细菌来源分子的反应、脂筏、膜微区、核受体活性,及类固醇激素受体活性等方面。KEGG主要富集在PI3K-Akt信号通路、AGE-RAGE信号通路、肿瘤坏死因子信号通路,及白细胞介素-17信号通路等。结论:黄芪桂枝五物汤通过多成分、多靶点及多途径的方式治疗类风湿关节炎。
Objective:To explore the molecular mechanism of Huangqi Guizhi Wuwu Tang(HGWT,黄芪桂枝五物汤)in the treatment of rheumatoid arthritis based on network pharmacology.Methods:Database TCMSP was used to search and screen the active components and targets of HGWT.GeneCards was used to predict the targets of rheumatoid arthritis,and the active components target network of traditional Chinese medicine was constructed.The obtained targets were introduced into the database STRING to construct protein-protein interaction network,and the target genes were enriched by GO and KEGG.Results:A total of 74 active components of HGWT were selected,which acted on 217 targets and 4329 targets of rheumatoid arthritis,thus 146 common targets of drugs and diseases were obtained.GO analysis involved the response to lipopolysaccharide,bacteria derived molecules,lipid rafts,membrane microdomains,nuclear receptor activity and steroid hormone receptor activity.KEGG is mainly enriched in PI3K-Akt signaling pathway,AGE-RAGE signaling pathway,tumor necrosis factor signaling pathway and interleukin-17 signaling pathway.Conclusion:HGWT can treat rheumatoid arthritis by multi-component,multi-target and multi-channel.
