Impairment of Lymphatic Microcirculation in Spontaneously Hypertensive Rats

摘要 目的:观察自发性高血压大鼠(SHRs)淋巴微循环功能变化。方法:8周龄雄性SHR大鼠(SHR组)和WKY大鼠(WKY组),每组各10只。应用VasTrack测量两组大鼠微淋巴管自律运动;取胸导管分离原代淋巴管内皮细胞(LECs)。应用免疫荧光和Western Blot检测LECs血管内皮生长因子受体3(VEGFR3)和淋巴管内皮透明质酸受体-1(LYVE-1)蛋白的表达;应用MTT法、基质胶成管和划痕实验检测两组LECs增殖、成管和迁移能力。结果:SHR组大鼠微淋巴管自律运动功能受损,相对振幅显著降低(P<0.01)。两组LECs LYVE-1、VEGFR3均呈阳性表达,且SHR组LECs VEGFR3蛋白表达水平显著性高于WKY组(P<0.01);SHR组LECs单视野内划痕细胞数显著低于WKY组(P<0.05);SHR组LECs管样结构数量显著增多(P<0.05)。结论:SHR组淋巴微循环功能受损,可能与LECs功能障碍有关。 Objective:To investigate the changes of functional status of lymphatic microcirculation in spontaneously hypertensive rats(SHRs).Method:The rats of experiments were divided into SHRs and normotensive control Wistar Kyoto rats(WKY)(n=10 in each group).Functional status of micro-lymphatic vasomotion of 8-week-old WKYs and SHRs were analyzed by a homemade VasTrack system.Thoracic ducts were taken from two groups of rats and primary lymphatic vessel endothelial cells(LECs)were isolated.Immunofluorescence and Western blot were applied to determine the expressions of vascular endothelial growth factor receptor 3(VEGFR3)and lymphatic vessel endothelial hyaluronic acid receptor 1(LYVE-1).The proliferation,tube-forming,and migration capabilities of LECs in two groups were evaluated and compared by using the MTT,matrix gel tube-forming,and wound scratch methods,respectively.Results:The functional status of micro-lymphatic vasomotion was impaired in the SHR group with decreased relative amplitude(P<0.01).LYVE-1 and VEGFR3 were positively expressed in both groups.The expression of VEGFR3 in SHRs LECs was significantly higher than that of WKYs(P<0.01).The migration capability of SHRs LECs was deteriorated with significantly decreased number of migrated cells in a single visual scratch field than that in the WKY group(P<0.05).Moreover,SHRs showed an increased number of formed tube-like structures(P<0.05).Conclusion:SHRs exhibited dysfunctional micro-lymphatic microcirculation,which might be related to dysfunctional LECs.
