摘要
本文建立了吗啡、可待因和6-单乙酰吗啡的毛细管电泳pH碱修饰在线富集方法,并进行了方法学考察。采用pH 3浓度为100mmol/L的磷酸盐缓冲液(含20%甲醇和5%异丙醇)作为背景缓冲液,0.2psi压力下引入3s含12.5%氨水的甲醇:水(50:50)混合溶液,10kV电压下电迁移进样99s,最后施加25kV电压进行分离检测。3种毒品在20-1000ng/mL范围内具有良好的线性关系,线性相关系数(r)均大于0.99,检出限均为7g/mL,相对标准偏差(RSD)在5.7%-12.2%(n=5)之间。该方法样品前处理简单,只需用甲醇稀释样品便可进样分析,方法灵敏度较高、稳定性较好,可用于唾液中吗啡类毒品的检验。
Base-mediated stacking in capillary electrophoresis were developed and validated in the present paper for morphine-type drugs analysis in saliva.Optimum conditions for the determination of morphine,codeine and 6-monoacetylmorphine in base-mediated stacking CE were investigated.100 mmol/L phosphate buffer(pH 3)containing 20%methanol and 5%isopropanol(v/v)was used as the background electrolyte for CE separation.The samples were introduced electrokinetically for 99 s at 10 kV in the stacking.12.5%aqueous ammonia and methanol(50:50,v/v)was introduced for 3 s under a pressure of 0.2 psi before sampling.The method showed a significant decrease in LODs(7 ng/mL),while providing good separation efficiency,and reproducibility(RSD:5.7~12.2%,n=5).Beyond this,sample pretreatment was simple,which only involved diluting with methanol.The method can be used to analyze morphine-type drugs in human saliva.
