摘要
目的了解癫痫持续状态(SE)后7 d海马各区巢蛋白(nestin)阳性星形胶质细胞表达及数目的变化,探讨其与SE导致海马区胶质瘢痕形成的关系。方法SD大鼠随机分为SE组和对照组,应用匹罗卡品建立SE大鼠模型;在SE后7 d处死大鼠行盲法免疫荧光组化实验,观察大鼠海马各区nestin与胶质纤维酸性蛋白单克隆抗体(GFAP)阳性细胞形态与数目变化及两者共标重叠情况,并进行统计学分析。结果SE组大鼠在给予匹罗卡品后均出现Ⅳ级以上持续状态癫痫发作。SE后7 d:SE组大鼠海马nestin阳性细胞数目显著增加,且广泛分布于海马各区,并与GFAP阳性细胞大量共标重叠;而对照组大鼠海马nestin染色阳性细胞只少量出现在海马齿状回颗粒层,与GFAP阳性细胞无共标重叠。结论SE后海马区大量表达的星形胶质细胞样nestin免疫阳性细胞可能是未分化成熟的星形胶质细胞,具有胚胎细胞的特性,可能与SE后海马区胶质瘢痕形成有关。
Objective To study the changes of numbers of Nestin positive astrocytes 7d after status epilepticus(SE)in the hippocampus,and to explore the relationship between Nestin positive astrocytes and SE in the hippocampus glial scar formation.Methods SD rats were randomly divided into SE group and the control group.SE was induced by injection of pilocarpine.the rats were sacrificed in 7 days after SE for the experiment of immunofluorescence histochemistry.Then the expressions of GFAP and Nestin in hippocampus of the two groups were observed and photographed under the confocal microscope.The number of positive cells was counted for statistical analysis.Results The number of Nestin positive cells in SE group were largely distributed in each hippocampal region,and were costandard overlap with GFAP positive cells.While in the control group,only a small amount of Nestin positive cells appeared in the DG of hippocampal,and there was no costandard overlap with GFAP positive cells.Conclusion The Nestin positive astrocytes expressed in the hippocampus after SE may be immature astrocytes,with the characteristics of embryonic cells,which may be related to the formation of glial scar in the hippocampus after SE.
