野蚕豆根总环烯醚萜苷的响应面优化提取工艺及抗氧化活性研究 被引量:4

摘要 以野蚕豆根为原料,优化总环烯醚萜苷的提取工艺。在单因素实验基础上,选取三因素三水平Box-Behnken实验设计,以总环烯醚萜苷得率为响应值,考察乙醇浓度、提取温度、液料比对总环烯醚萜苷提取效果的影响;并对野蚕豆根中总环烯醚萜苷进行了抗氧化的体外活性实验。结果表明:提取野蚕豆根中总环烯醚萜苷的最佳条件为:乙醇浓度60%、提取温度50℃、液料比30∶1mL/g,此条件下重复实验,总环烯醚萜苷得率为2.425%,RSD为1.064%。在抗氧化实验中,总环烯醚萜苷对DPPH自由基、ABTS+自由基和羟基自由基有一定的清除能力,其清除DPPH自由基、ABTS+自由基和羟基自由基的半抑制浓度(IC50)分别为:0.078mg/mL、0.1397mg/mL和1.15mg/mL。 This study aimed to optimize the extraction process of total iridoid glycoside with Centranthera grandiflora as raw material.On the basis of single factor experiment,three factors and three levels Box-Be hnken experimental design was selected to investigate the effects of ethanol concentration,extraction temperature and liquid-solid ratio on the extraction efficiency of total iridoid glycosides.The antioxidant activity of total iridoid glycoside in Centranthera grandiflora was then studied in vitro.The results showed that the best conditions for extraction process were:ethanol concentration 60%,extraction temperature 50℃,solid-liquid ratio 30∶1(mL/g).Under these conditions,the content of total iridoid glycoside was 2.425%with a relative standard error(RSD)of 1.064%.In the antioxidant experiment,the total iridoid glycoside possessed free radical scavenging capacity,including DPPH free radical,ABTS+free radical and hydroxyl free radical with a semi-inhibitory concentration(IC50)of 0.078 mg/mL,0.1397 mg/mL and 1.15 mg/mL,respectively.
