摘要
为比较鸡传染性囊病(IBD)免疫复合物疫苗和活疫苗免疫对鸡外周血免疫淋巴细胞功能的影响,本试验用IBD免疫复合物疫苗和BX株活疫苗免疫1日龄SPF鸡,并设立空白对照,于免疫后7、14、21、2835 d和42 d采用流式细胞仪检测淋巴细胞凋亡率及CD4^+T、CD8^+T、Bu-1^+B细胞数变化。结果显示,免疫后7 d,活疫苗组淋巴细胞凋亡率极显著高于免疫复合物疫苗组和对照组(P<0.01),其他时间3组均无显著性差异(P>0.05);与对照组相比,免疫复合物疫苗组免疫后7~42 d主要引起CD4^+T细胞数升高(P<0.05或P<0.01),活疫苗组主要引起CD8^+T细胞数升高(P<0.05或P<0.01),但均能导致Bu-1^+B细胞数减少(P<0.05或P<0.01)。表明本免疫复合物疫苗和活疫苗均能增强鸡外周血细胞免疫功能,但对体液免疫均有一定的抑制作用。本试验为IBD免疫复合物疫苗的研发提供了依据。
To compare the effection on immune function in chicken peripheral blood vaccinated with infectious bursal disease immune complex vaccine(IBD IC)and live vaccine.One-day-old SPF chickens were vaccinated with IBD IC vaccine and BX strain live vaccine,and blank control group was set up.The apoptosis rate and changes of CD4^+T,CD8^+ T and Bu-1^+B lymphocytes in peripheral blood of chickens were measured by flow cytometry on days 7,14,21,28,35,and 42.The results showed that the apoptosis rate of peripheral blood lymphocytes in live vaccine group was significantly higher than that of IBD IC vaccine group and control group on 7 days after immunization(P<0.01),but no difference between the three groups at other times(P>0.05).Compared with the control group,the number of CD4^+T cells increased mainly after vaccination with IBD IC vaccine(P<0.05 or P<0.01)and CD8^+T cells increased mainly after vaccination with live vaccine from 7 to 42 days(P<0.05 or P<0.01),but all of them lead to a decrease in the number of Bu-1^+B cells(P<0.05 or P<0.01).The results showed that both IBD IC and live vaccine could enhance the cellular immune function,but had inhibitory effects on humoral immunity.
