可溶性CD40配体对非霍奇金淋巴瘤raji细胞凋亡及PD-L1影响

Effect of soluble CD40 ligand on apoptosis and PD-L1 in non-Hodgkin’s lymphoma raji cells

摘要 目的分析可溶性CD40配体(sCD40L)对非霍奇金淋巴瘤(non-Hodgkinlymphoma,NHL)raji细胞增殖凋亡及凋亡死亡受体-配体1(PD-L1)的影响。方法实验组(加入2、4、6μg/mL的sCD40L)和对照组(未加sCD40L)作用NHL raji细胞24h,采用CCK-8法检测细胞增殖,流式细胞术AnnexinⅤFITC/PI法检测细胞凋亡,采用免疫细胞化学(immunocytochemistry,ICC)法检测PD-L1的表达。结果2、4、6μg/mL的sCD40L作用NHL raji细胞24h后,细胞增殖抑制率分别为(27.59±2.39)%、(41.96±1.76)%和(72.58±2.64)%,与对照组(11.86±1.53)%比较(线性F=1310.57,P<0.001),线性回归分量有统计学意义,说明存在相关关系即有线性趋势;线性偏差F=23.83,P<0.001,偏离线性回归分量有统计学意义,说明不是简单的线性关系。细胞凋亡率分别为(12.36±2.04)%、(28.61±3.73)%和(35.80±4.16)%,与对照组(3.53±0.37)%比较(线性F=215.94,P<0.001),线性回归分量有统计学意义,说明存在相关关系即有线性趋势;线性偏差F=2.41,P=0.152,偏离线性回归分量无统计学意义,说明是简单的线性关系。PD-L1表达分别为(81.00±3.00)%、(70.00±4.00)%和(12.00±2.00)%,与对照组(93.00±4.00)%比较(线性F=860.21,P<0.001),线性回归分量有统计学意义,说明存在相关关系即有线性趋势;线性偏差F=85.89,P<0.001,偏离线性回归分量有统计学意义,说明不是简单的线性关系。结论不同浓度的sCD40L可以抑制NHL raji细胞增殖、诱导凋亡,其机制可能与降低PD-L1表达有关。 OBJECTIVE To investigate the effects of soluble CD40 ligand(sCD40 L)on Non-Hodgkin’s lymphoma(NHL)raji cell proliferation and apoptosis and programmed death ligand 1(PD-L1,also known as CD274,B7-H1).METHODS The experimental group(the concentration of sCD40 L was 2,4 and 6μg/ml)and the control group(no sCD40 Lwas added)was used to treat non-Hodgkin’s lymphoma raji cells for 24 hours.Cell proliferation was detected by CCK-8 method.Apoptosis was detected by flow cytometry AnnexinⅤFITC/PI method.Immunocytochemistry(ICC)method was used to detect the expression of PD-L1.RESULTS After treating Raji cells with 2,4 and 6μg/ml of sCD40 L for 24 h,our data showed that cell proliferation inhibition rates were(27.59±2.39)%,(41.96±1.76)%and(72.58±2.64)%,compared with the control group(11.86±1.53)%(F=1310.57,P<0.001),the linear regression component had statistical significance(F=23.83,P<0.001),and deviation from linear regression component had statistical significance,indicating that it was not a simple linear relationship.The apoptosis rates were(12.36±2.04)%,(28.61±3.73)%and(35.80±4.16)%,compared with the control group(3.53±0.37)%(F=215.94,P<0.001),the linear regression component had statistical significance,indicating that there was a linear trend when there was a correlation.Deviation from linear regression component had no statistical significance(F=2.41,P=0.152),indicating a simple linear relationship.The expressions of PD-L1 were(81.00±3.00)%,(70.00±4.00)%,(12.00±2.00)%,compared with the control group(93.00±4.00)%(F=860.21,P<0.001),linear regression the weight was statistically significant,indicating that there was a linear trend when there is a correlation.Deviation from linear regression component had statistical significance(F=85.89,P<0.001),indicating that was not a simple linear relationship.CONCLUSION Different concentrations of sCD40 Lcan inhibit the proliferation and induce the apoptosis of NHL raji cells,which may be related to the reduction of PD-L1 expression.
