摘要
目的鉴定食管鳞癌发生发展过程中的核心基因,寻找新的分子靶标。方法 GEO数据库筛选出食管鳞癌数据集GSE20347,GSE20344,GEO2R分析差异基因。利用STRING和Cytoscape建立并修饰蛋白与蛋白相互作用网络,GO功能和KEGG通路富集分析。Cytohubba计算核心基因,TCGA数据验证核心基因的表达情况。Kaplan Meier-plotter数据库用于核心基因的生存分析。结果获得共同上调基因19个和下调基因39个。其主要参与细胞外基质组织,定位于细胞外区域部分,功能为丝氨酸肽酶活性,涉及蛋白质消化吸收相关通路。12个核心基因在TCGA数据集的表达基本一致。CDH11,VCAN和SPP1表达与食管鳞癌预后显著相关。结论 CDH11和VCAN的下调以及SPP1的过表达被确定为食管鳞癌不良预后因素,为食管鳞癌的诊断和预后提供了新的标志物。
Objective To identify the hub genes in the development of esophageal squamous cell carcinoma and find new molecular targets. Methods Data sets GSE20347, GSE20344 and GEO2 R of esophageal squamous cell carcinoma were screened from geo database. The protein-protein interaction network was established and modified by STRING and Cytoscape, and the GO function and KEGG pathway enrichment were analyzed. Cytohubba was used to calculate core genes, and TCGA data was used to verify the expression of core genes. Kaplan Meier plotter database was used for survival analysis of core genes. Results 19 up-regulated genes and 39 down regulated genes were obtained. It is mainly involved in extracellular matrix tissue, located in the extracellular region, and its function is serine peptidase activity, involving in protein digestion and absorption related pathways. The expression of 12 core genes in TCGA data set was basically consistent. The expressions of CDH11, VCAN and SPP1 were significantly correlated with the prognosis of ESCC.Conclusions The down-regulation of CDH11 and VCAN and the overexpression of SPP1 are identified as poor prognostic factors of ESCC, which provide new markers for the diagnosis and prognosis of ESCC.
