褪黑素对大鼠挤压综合征模型骨骼肌缺血再灌注损伤的影响 被引量:2

摘要 目的观察褪黑素对大鼠挤压综合征模型骨骼肌缺血再灌注损伤的影响。方法将wistar雄性大鼠随机分为空白组(Sham组)、对照组(CS组)、氯化钠组(NS组)及褪黑素组(MEL组),给予5%戊巴比妥钠腹腔注射,麻醉大鼠后,挤压大鼠后肢5 h,随后再灌注6 h。MEL组和NS组造模后在解压即刻分别静脉注射10 mL/kg褪黑素溶液与等体积的生理盐水。收集各组大鼠后肢再灌注后6 h血清及腓肠肌样品用于生化分析及组织学评估,同时观察生存率。结果组织缺血再灌注过程中,CS组血清中钾离子、CK、LDH含量较Sham组显著增加(P<0.05);与CS组相比,MEL组血清中钾离子、CK、LDH的含量显著降低(P<0.05)。同样,与Sham组相比,CS组血清中GSH-Px、SOD、CAT的含量显著降低,MDA的含量显著增加(P<0.05);与CS组相比,MEL组血清中GSH-Px、SOD、CAT的含量显著升高,MDA含量显著降低(P<0.05)。光镜下腓肠肌HE染色显示不同程度组织破坏及间质水肿,MEL组腓肠肌缺血再灌注损伤得到明显改善。CS组与NS组大鼠存活率显著降低,MEL组大鼠存活率高于CS组与NS组(P<0.05)。结论褪黑素对挤压综合征骨骼肌缺血再灌注损伤具有保护作用,并能提高挤压综合征模型大鼠的生存率。 Objective To observe the effect of Melatonin on skeletal muscle ischemia-reperfusion injury in rat model of crush syndrome.Methods Male wistar rats were randomly divided into Sham group,CS group,NS group and MEL group.5%sodium pentobarbital was injected intraperitoneally,The rats were anesthetized and the hind limbs were compressed for 5 h,and then reperfused for 6 h.Immediately after decompression,10 ml/kg melatonin solution and the same volume of saline were injected intravenously in the MEL group and the NS group.The serum and gastrocnemius muscle samples were collected for biochemical analysis and histological evaluation,and the survival rate was observed.Results The contents of K+,CK and LDH in serum of CS group were significantly higher than those of Sham group(P<0.05),and the contents of K+,CK and LDH in serum of MEL group were significantly lower than those of CS group(P<0.05).Similarly,compared with Sham group,the contents of GSH-Px,SOD,CAT in CS group were significantly decreased,the contents of MDA were significantly increased(P<0.05),the contents of GSH-Px,SOD,CAT in Mel group were significantly increased,and the contents of MDA were significantly decreased(P<0.05).Under light microscope,HE staining of gastrocnemius showed different degree of tissue destruction and interstitial edema,MEL group gastrocnemius ischemia-reperfusion injury was significantly improved.The survival rate of CS group and NS group was significantly lower than that of MEL group(P<0.05).Conclusion Melatonin has protective effect on skeletal muscle ischemia-reperfusion injury in crush syndrome rats and can improve the survival rate of crush syndrome rats.
