Complexin蛋白通过C端序列加速突触形成

摘要 为探究complexin对小鼠大脑皮层神经元生长的影响,将过表达的complexin及突变体导入小鼠大脑皮层神经元.利用共聚焦显微镜对神经元进行免疫荧光成像并利用Image J对突触的密度及平均大小进行分析.结果表明:过表达complexin后,突触密度上升而平均大小无明显变化;去除complexin的C末端后,能明显抑制由其带来的突触密度上升,但去除N末端却没有相同的效果;改变complexin的定位与过表达complexin的作用相同.说明complexin能促进小鼠大脑皮层神经元的生长,这种功能可能来自于它的C末端本身,与complexin的定位无关. To investigate the effects of complexin on the growth of cortical neurons in mice,complexin and its mutants were overexpressed on cortical neurons of mice.The growth of neurons was monitored by confocal microscope by immunofluorescence imaging,and analyzed by using Image J to determine the density and average size.The results showed after overexpression of complexin,the synaptic density increased but the average size did not change;after removal of C-terminal,the increase of synaptic density was inhibited,but the removal of N-terminal did not show the same effect;the effect of the location of complexin was the same as the complexin overexpression.So complexin could promote the growth of cortical neurons in mice which was a function that probably came from its C-terminal itself,regardless of where the complexin was located.
