miR-24-3p在肺结核病患者血清中的表达及其临床意义 被引量:3

摘要 目的探讨miR-24-3p在肺结核病患者血清中的表达及其作为分子标志物的诊断价值。方法选取在新疆喀什地区第一人民医院就诊的肺结核病患者50例为病例组(肺结核病组),其中活动性肺结核30例,非活动性肺结核20例;其他肺部疾病患者30例及健康体检者30例为健康对照组,采用qRT-PCR法测定血清中miR-24-3p的相对表达量,并采用ROC曲线评价miR-24-3p在肺结核病诊断及鉴别诊断中的价值。结果50例肺结核患者血清中miR-24-3p的表达量明显低于健康对照组和其他肺部疾病组(P均<0.01);其他肺部疾病组的表达量低于健康对照组(P<0.01);活动性肺结核组低于非活动性肺结核组(P<0.01)。ROC曲线分析表明miR-24-3p对于区分肺结核组与健康对照组的曲线下面积(AUC)为0.9267[95%CI(0.8637-0.9896)],灵敏度和特异度分别为86%和90%;区分肺结核病组和其他肺部疾病组的AUC为0.7260,95%CI(0.6166-0.8354),灵敏度和特异度分别为64%和83%;miR-24-3p对于区分活动性肺结核与非活动性肺结核的曲线下面积(AUC)为0.7725,95%CI(0.6425-0.9025),灵敏度和特异度分别为53.3%和100%。结论miR-24-3p在肺结核患者血清中低表达,其可作为一种潜在标志物,对结核病的诊断及活动性鉴别具有一定价值。 Objective To investigate the expression of miR-24-3p in the serum of patients with pulmonary tuberculosis and its diagnostic value as a molecular marker.Methods 50 cases of pulmonary tuberculosis patients admitted to the First People's Hospital of Kashgar were selected as case group,among which 30 patients of active tuberculosis and 20 patients of inactive tuberculosis were included,and 30 patients with other pulmonary diseases and 30 healthy physical examination subjects were the control group.The relative expression of miR-24-3p in serum was measured by qRTPCR,and the value of miR-24-3p for diagnosis and differential diagnosis of tuberculosis was evaluated by ROC curve.Results The expression level of miR-24-3p in serum of 50 patients with pulmonary tuberculosis was significantly low⁃er than that in the healthy control group and other pulmonary diseases group(both P<0.01);The expression level in other pulmonary diseases group was lower than that of healthy control group(P<0.01);The expression level in active pulmonary tuberculosis was lower than that of inactive pulmonary tuberculosis(P<0.01).ROC curve analysis showed that the area under the curve(AUC)of miR-24-3p for distinguishing pulmonary tuberculosis from the healthy controls was 0.9267[95%CI(0.8637-0.9896)],and the sensitivity and specificity was 86%and 90%,respectively;The AUC of the pulmonary tuberculosis and other lung diseases was 0.7260[95%CI(0.6166-0.8354)],and the sensitivity and spec⁃ificity was 64%and 83%;The AUC of miR-24-3p for distinguishing active tuberculosis from inactive tuberculosis was 0.7725[95%CI(0.6425-0.9025)],and the sensitivity and specificity was 53.3%and 100%,respectively.Conclu⁃sion miR-24-3p is low expressed in the serum of patients with pulmonary tuberculosis,which can be used as a poten⁃tial marker and has a certain value for the diagnosis and activity of tuberculosis.
