葛根素通过抑制IκBα/NF-κB p65信号轴缓解大鼠非酒精性脂肪肝损伤 被引量:7

Puerarin Alleviates Nonalcoholic Fatty Liver Disease-Induced Liver Injury in Rats by Inhibiting IκBα/NF-κB p65 Signaling Axis

摘要 【目的】观察葛根素对大鼠非酒精性脂肪肝(NAFLD)的治疗作用及机制。【方法】将60只大鼠随机分为正常对照组,模型对照组,葛根素低、中、高剂量组,多烯磷脂酰胆碱组,每组10只。除正常对照组外,其余组别大鼠给予高脂饲料构建NAFLD模型。造模同时,葛根素低、中、高剂量组分别给予葛根素10、50、100 mg·kg-1·d-1灌胃,多烯磷脂酰胆碱组给予多烯磷脂酰胆碱50 mg·kg-1·d-1灌胃,正常对照组和模型对照组给予等容量生理盐水灌胃。连续12周。以苏木素-伊红(HE)染色法观察大鼠肝脏组织病理变化,全自动生化仪检测血清谷氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白(LDL)、高密度脂蛋白(HDL)水平,酶联免疫吸附分析(EILSA)检测外周血肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)、诱导型一氧化氮合酶(iNOS)水平,油红O染色法观察肝脏中脂肪沉积情况,蛋白免疫印迹法检测肝组织Caspase-3、Caspase-9、核因子(NF)κB抑制剂α(IκBα)、NF-κB p65活化水平。【结果】与正常对照组比较,模型对照组大鼠肝脏组织可见结构损坏、过多脂肪沉积,血清ALT、AST、TG、TC、LDL、TNF-α、IL-6、iNOS含量均显著升高(P <0.05),HDL含量显著降低(P <0.05),肝组织Caspase-3、Caspase-9相对表达量,p-IκBα/IκBα、p-NF-κB p65/NF-κB p65比值均显著升高(P <0.05);与模型对照组比较,葛根素中、高剂量组和多烯磷脂酰胆碱组可显著改善上述指标水平(P <0.05),且葛根素高剂量组上述指标水平与多烯磷脂酰胆碱组比较,差异无统计学意义(P> 0.05)。【结论】葛根素可缓解NAFLD大鼠肝脏损伤,降低血脂水平,其机制可能与抑制IκBα/NF-κB p65信号轴有关。 Objective To observe the therapeutic effects of puerarin on nonalcoholic fatty liver disease(NAFLD)in rats and its mechanism. Methods Sixty SD rats were randomly divided into normal control group,model control group,low-,middle-,high-dose puerarin groups,and polyene phosphatidyicholine(PPC)group,10 rats in each group. Apart from the normal control group,the rats in the other groups were induced into NAFLD model by feeding high-lipid fodder. After successful modeling,the low-,middle-,high-dose puerarin groups were given intragastric administration of 10, 50, 100 mg · kg-1· d-1 of puerarin, respectively, the PPC group was given intragastric administration of 50 mg·kg-1·d-1 of PPC,and the normal control group and model control group were given intragastric administration of the same volume of normal saline, the treatment lasting 12 weeks. After medication,the morphological features of liver tissue were observed under microscope by hematoxylin-eosin(HE)staining,the serum levels of alanine aminotransferase(ALT),aspartate transaminase(AST),triglyceride(TG),total cholesterol(TC),low density lipoprotein(LDL),and high density lipoprotein(HDL)were determined by automatic biochemical analyzer,the serum levels of tumor necrosis factor α(TNF-α),interleukin 6(IL-6)and inducible nitric oxide synthase(iNOS)were detected by enzyme-linked immunosorbent assay(ELISA),and the lipid deposition in liver was observed by oil red O staining,and the expression levels of Caspase-3,Caspase-9 and activation levels of inhibitor of nuclear factor(NF)-κB α(IκBα)and NF-κB p65 in liver tissue were detected by Western blotting assay. Results Compared with the normal control group,the damaged structure and more fat deposition in liver tissue were seen in the model control group,the serum contents of ALT,AST,TG,TC,LDL,TNF-α,IL-6,iNOS were increased(P < 0.05),serum HDL content was decreased significantly(P <0.05),and the relative expression levels of Caspase-3,Caspase-9,and ratio of p-IκBα/IκBα and p-NF-κB p65/NF-κB p65 in liver tiss
