摘要
目的:建立高效液相色谱法测定大鼠血浆中苍耳亭的浓度。方法:采用高效液相色谱法,色谱柱为Hypersil ODS2柱(250 mm×4.6 mm,5μm),流动相为甲醇-水(V∶V=52∶48),流速为1.0 ml/min,检测波长为278 nm,柱温为25℃。血浆样品经二氯甲烷萃取富集,10μl进样检测。结果:苍耳亭质量浓度在0.1~10μg/ml(R^2=0.9997)范围内呈现良好的线性关系,定量下限为0.1μg/ml。苍耳亭低、中及高浓度的提取回收率分别为89.19%、83.56%及96.72%,日内和日间精密度分别≤5.47%和8.65%。结论:本方法灵敏快速简单,适用于体内苍耳亭的浓度分析,为药动学研究提供方法依据。
OBJECTIVE:To establish the high performance liquid chromatography(HPLC)method for determination of concentration of xanthine in rat plasma.METHODS:HPLC methods was adopted with Hypersil ODS2 column(250 mm×4.6 mm,5μm).The mobile phase was methanol-water(V∶V=52∶48),the flow rate was 1.0 ml/min,the detection wavelength was 278 nm,and the column temperature was 25℃.The plasma samples were extracted and enriched with dichloromethane,and 10μl injection was used for detection.RESULTS:The calibration curve of xanthatin in plasma showed good linear correlation in the range from 0.1 to 10μg/ml(R^2=0.9997)and the lower limit of concentration was 0.1μg/ml.The extraction recovery rates of low,medium and high concentration of Xanthatin was respectively 89.19%,83.56%and 96.72%,and the intra-day and inter-day precision was respectively≤5.47%and 8.65%.CONCLUSIONS:This method is sensitive,rapid and simple,and is suitable for the analysis of the concentration of xanthine in vivo,and provides basis for pharmacokinetic research.
