摘要
目的采用网络药理学方法探索三七化瘀止血功能在心房颤动(AF)治疗中维持抗凝/出血稳态的机理。方法在中药系统药理学数据库与分析平台(TCMSP)、中国天然产物化学成分库等数据库检索三七相关中药化学成分及潜在靶点,通过GeneCards、OMIM数据库挖掘抗凝/出血相关的作用靶点,两者取交集筛选,再进行富集分析。结果筛选出三七生物活性成分8种,预测出三七相关的靶点189个,抗凝/出血相关的靶点3078个。通过韦恩网取交集得到三七在维持抗凝/出血稳态的共同作用靶点(关键靶点)98个。DAVID富集分析显示,三七维持抗凝/出血稳态主要涉及血小板激活通路、Jak-STAT信号通路、Wnt信号通路、VEGF信号通路、癌症通路、TNF信号通路、PI3K-Akt信号通路、甲状腺激素信号通路、Ras信号通路等107条通路。结论三七化瘀止血功能在AF治疗中维持抗凝/出血稳态主要涉及血小板调节作用,其他还与抗肿瘤、抗动脉粥样硬化、调节内分泌、调节免疫、抗炎等多靶点、多通路相关,体现出中医药多环节发挥作用的特点。
Objective To explore the mechanism of panax notoginseng's eliminating blood stasis and hemostasis in maintaining anticoagulation/bleeding homeostasis in the treatment of atrial fibrillation(AF)by network pharmacology.Methods The chemical components and potential targets of panax notoginseng were searched in the databases of traditional Chinese medicine by systems platform(TCMSP)and chemical sublibrary of Chinese natural products,etc.The targets related to anticoagulation/bleeding were mined by Genecards and OMIM database.The intersection was selected for screening and enrichment analysis.Results Eight bioactive components of panax notoginseng were screened,189 targets related to panax notoginseng and 3078 targets related to anticoagulation/hemorrhage were predicted.A total of 98 common action targets(key targets)of panax notoginseng in maintaining anticoagulation/bleeding homeostasis were obtained by Veen net.DAVID enrichment analysis showed that 107 pathways were involved in the maintenance of anticoagulation/hemorrhage homeostasis of panax notoginseng,including platelet activation pathway,Jak-STAT signal pathway,Wnt signal pathway,VEGF signal pathway,cancer pathway,TNF signal pathway,PI3K-Akt signal pathway,thyroid hormone signal pathway,Ras signal pathway and so on.Conclusion Panax notoginseng's eliminating blood stasis and hemostasis in maintaining anticoagulation/bleeding homeostasis in the treatment of AF mainly involves platelet regulation.Others are also related to anti-tumor,anti-atherosclerosis,regulation of endocrine,regulation of immunity,anti-inflammation and other multi-targets and multi-pathways.The study reflects the characteristics of the role of traditional Chinese medicine in multiple links.
