全身交变电磁辐照疗法对2型糖尿病小鼠神经病理性疼痛的作用效果研究

摘要 目的:观察全身交变电磁辐照对2型糖尿病(db/db)小鼠神经病理性疼痛的改善效果。方法:将20只3月龄雄性2型糖尿病(db/db)小鼠及10只同月龄、同背景的野生型小鼠分成3组:野生型小鼠组(wild-type组)、db/db小鼠组(db/db组)、db/db小鼠接受交变电磁辐照(alternating electromagnetic fields,EMF)组(db/db+EMF组)。对db/db+EMF组小鼠施加1 h、连续4周的全身交变电磁辐照,对wild-type组和db/db组小鼠施加假电磁干预。分别于治疗的第0、1、2、3、4周检测各组小鼠足底机械痛阈值、热痛阈值、感觉神经传导速率、脊髓p38丝裂原活化蛋白激酶(p38 MAPK)表达和脊髓白细胞介素-1β(IL-1β)表达。结果:db/db+EMF组小鼠在治疗的第1、2、3、4周足底机械痛阈值、热痛阈值和感觉神经传导速率均显著高于db/db组小鼠(P<0.05),并且db/db+EMF组小鼠脊髓p38 MAPK和IL-1β的基因表达水平显著低于db/db组小鼠(P<0.05)。结论:全身交变电磁辐照能够显著缓解2型糖尿病(db/db)小鼠神经病理性疼痛症状,具有潜在的临床应用前景。 Objective To observe the ameliorative effects of whole-body alternating electromagnetic exposure on neuropathic pain in type 2 diabetic(db/db)mice.Methods Twenty 3-month-old male type 2 diabetic(db/db)mice and 10 wild-type mice with the same age and background were divided into 3 groups:a wild-type group(wild-type),a db/db mice group(db/db)and a db/db mice with alternating electromagnetic exposure(db/db+EMF)group.The mice in the db/db+EMF group were subjected to 1 h of whole-body alternating electromagnetic irradiation for 4 weeks,and the mice in the wild-type and db/db groups went through sham electromagnetic intervention.The mice in the wild-type group and the db/db group were subjected to the sham alternating electromagnetic exposure.The paw mechanical pain threshold,thermal pain threshold,sensory nerve conduction velocity,p38 mitogen-activated protein kinase(p38 mitogen-activated protein kinase,p38 MAPK)and interleukin-1β(interleukin-1β,IL-1β)expression were determined at 0,1,2,3,and 4 weeks after the laser treatment.Results The mice in the db/db+EMF group exhibited significantly higher paw mechanical pain threshold,thermal pain threshold and sensory nerve conduction velocity than the mice in the db/db group at 0,1,2,3,and 4 weeks after treatment(P<0.05).Moreover,the mice in the db/db+EMF group showed significantly lower spinal p38 MAPK and IL-1βexpression levels than the mice in the db/db group(P<0.05).Conclusion Whole-body alternating electromagnetic exposure has the capacity to attenuate neuropathic pain in type 2 diabetic(db/db)mice,and may be applied clinically in the future.[Chinese Medical Equipment Journal,2021,42(1):9-13,31]
