摘要
目的:基于网络药理学探讨生脉散治疗心律失常的分子机制。方法:使用TCMSP和TCMID数据平台筛选生脉散的有效成分,TCMSP及STITCH数据库筛选药物作用靶点,通过TTD、DRUGBANK及DisGeNET数据库检索心律失常的疾病靶点。制作韦恩图获得相关交集,使用String数据库制作靶点间相互作用网络,并依据置信度筛选关键靶点及关键成分,运用DAVID网站进行KEGG和GO通路富集分析,使用Cytoscape软件构建成分-靶点-通路网络。结果:最终筛选出生脉散关键成分19个,疾病靶点180个,作用于心律失常靶点23个,重要通路35条,KEGG和GO通路分析与筛选显示生脉散对心律失常的治疗作用或通过调节钙离子信号通路、腺苷酸环化酶激活肾上腺素能受体信号通路、心肌细胞的肾上腺素能信号等实现。结论:生脉散具有多成分、多靶点、多通路的作用特点,通过交感神经反射、离子通道、cGMP-PKG、cAMP等信号转导发挥治疗心律失常的作用。
Objective:To discuss the molecular mechanism of Shengmai Powder in treating arrhythmia based on network pharmacology.Methods:Use TCMSP and TCMID data platforms to screen the active ingredients of Shengmai Powder,TCMSP and STITCH databases to screen drug targets,and TTD,DRUGBANK and DisGeNET databases to search for arrhythmia disease targets.Make Venn diagrams to obtain relevant intersections,use String database to make interaction networks between targets,and screen key targets and key components by setting the confidence level,use DAVID website for KEGG and GO pathway enrichment analysis,and use Cytoscape software to build components-target-access network.Results:In the end,19 key ingredients of Shengmai Powder,180 disease targets,23 targets for arrhythmia,35 important pathways were screened.The analysis and screening of KEGG and GO pathways showed that Shengmai Powder has a therapeutic effect on arrhythmia or through regulation Calcium ion signal pathway,adenylate cyclase activation of adrenergic receptor signal pathway,and adrenergic signal of cardiomyocytes are realized.Conclusion:Shengmai Powder has the characteristics of multi-component,multi-target,and multi-pathway.It can treat arrhythmia through sympathetic nerve reflex,ion channels,cGMP-PKG,cAMP and other signal transduction.
