摘要
本研究为了筛选出扁桃斑鸠菊的抗乳腺活性部位,以人乳腺癌细胞株(MCF-7)为实验模型,采用CCK-8实验评价扁桃斑鸠菊提取物对乳腺癌细胞增殖的抑制作用。选择极性不同的溶剂,设计了三种不同的提取路线,最终得到极性不同的各个部位提取物(VA1-VA8)。大量提取活性较好的乙酸乙酯提取物,然后洗脱分离为6个段(A2-A7),继续运用CCK-8细胞毒性实验来追踪抗乳腺癌活性最佳的部位。CCK-8实验结果显示:扁桃斑鸠菊乙酸乙酯提取物对MCF-7细胞增殖表现出良好的抑制作用,且呈剂量依赖性。在A2-A7的6个洗脱产物中,浓度为100μg/mL时,50%乙酸乙酯石油醚洗脱段(A5)对人乳腺癌细胞MCF-7具有最佳的抑制效果,抑制率为83.34%。
The objective of the present study was to screen the extracts for anti-breast cancer activity from Vernonia amygdalina using human breast cancer cell lines(MCF-7)as a test model.CCK-8 assay was used to evaluate the inhibitory effect of extractsfrom V.amygdalina on the proliferation of breast cancer ceus.Three different methods were applied to extract the leaves of V.amygdalina with several solvents by different polarity and get several crude extracts(VA1-VA8).For the ethyl acetate extracts with better activities than others,a large number of V.amygdalina were extracted with ethyl acetate and divided into six sections(A2-A7),and the CCK-8 cytotoxic test was used to track the site again which had the best anti-breast cancer activity.Results from CCK-8 assay revealed that ethyl acetate extracts from V.amygdalina showed high activity against MCF-7 in a dose-dependent manner.A5 which was eluted by 50%EA-PE.had the best anti-breast cancer activity.The inhibition rate of A5 was 83.34%when its concentration was 100μg/mL.
