摘要
目的基于脑胶质瘤患者和健康志愿者血浆外泌体中N-糖基化蛋白质的表达差异,筛选潜在的药物靶标蛋白。方法分别取3名脑胶质瘤患者和3名健康志愿者的混合血浆并提取外泌体。基于肼化学法富集上述血浆外泌体中的N-糖基化蛋白质并对其进行质谱鉴定。比较脑胶质瘤患者和健康志愿者血浆外泌体N-糖基化蛋白质的表达差异,并确定潜在的药物靶标蛋白。结果脑胶质瘤患者和健康志愿者血浆外泌体中共有44个N-糖基化位点的表达量存在差异,归属于39个N-糖基化蛋白质。其中,7个N-糖基化蛋白质为潜在的药物靶标蛋白。结论脑胶质瘤患者血浆外泌体表达的N-糖基化蛋白质中含有潜在的药物靶标蛋白,为新药筛选及先导化合物开发与设计等奠定分子基础。
Objective Based on the different expression of N-glycoprotein between patients with glioma and healthy volunteers, potential protein drug targets were screened. Methods Mixed plasma were taken from three patients with glioma and three healthy volunteers and the exosomes were extracted, respectively. The N-glycoproteins in plasma exosomes were enriched by hydrazide chemical method and the obtained N-glycproteins were detected by mass spectrometry. Label free based quantitative method was used to compare the expressions of N-glycoprotein in plasma exosomes between patients with glioma and healthy volunteers and the potential protein drug targets were identified. Results Forty-four N-glycosylation sites were differently expressed in plasma exosomes of patients with glioma and healthy volunteers, which belong to thirty-nine N-glycosylation proteins. Among them, seven N-glycoproteins are potential protein drug targets. Conclusion N-glycoproteins which differently expressed in exosomes of patients with glioma contain potential protein drug targets, which is especially beneficial for new drug and leading compound development.
