ANP对胃癌MGC-803细胞增殖迁移的影响及机制的研究 被引量:5

摘要 目的:探讨心房钠尿肽(atrial natriuretic peptide,ANP)对人胃癌细胞株MGC-803的体外作用和联合PI3K/Akt信号通路抑制剂(LY294002)对人胃癌细胞株MGC-803的体外作用及其机制。方法:CCK8法分别检测不同浓度ANP和不同浓度LY294002对胃癌细胞MGC-803增殖的影响;MGC-803细胞随机分为四组:对照组、ANP组、LY294002组及ANP联合LY294002组,CCK8法检测细胞增殖情况;划痕实验检测细胞迁移;倒置显微镜下观察细胞形态的变化;Western blot检测PI3K/Akt通路中关键信号分子总Akt(t-Akt)、p-Akt蛋白的表达情况。结果:CCK8显示,ANP单用、LY294002单用对人胃癌细胞MGC-803增殖的抑制呈时间及剂量依赖性;与对照、ANP单用和LY294002单用比较,ANP与LY294002联合应用可显著抑制人胃癌细胞MGC-803的增殖,差异均有统计学意义(P<0.05)。划痕结果显示,对照组、ANP组、LY294002组、ANP+LY294002组24h划痕面积修复率依次为(50.99±1.80)%、(43.33±2.22)%、(35.81±4.09)%、(26.67±1.69)%,差异均有统计学意义(P<0.05)。四组MGC-803细胞分别出现不同的细胞形态学改变。Western blot结果显示,ANP和LY294002均可抑制MGC-803细胞中p-Akt蛋白的表达,且ANP联合LY294002比二者单独应用对p-Akt蛋白抑制表达作用更显著(P<0.05),而t-Akt表达未发生显著的改变(P>0.05)。结论:ANP可抑制人胃癌细胞MGC-803的增殖和迁移,与LY294002联用能增加对胃癌细胞MGC-803的抑制作用,其作用机制可能与下调PI3K/Akt信号通路活性有关。 Objective:To investigate the in vitro effect of atrial natriuretic peptide(ANP)on human gastric cancer cell line MGC-803 and the combined effect of PI3K/Akt signaling pathway inhibitor(LY294002)on human gastric cancer cell line MGC-803 and the possible mechanism.Methods:The gastric cancer MGC-803 cells were treated with different concentrations of ANP and different concentrations of LY294002.The cell proliferation inhibition rate was detected by MTT assay.MGC-803 cells were randomly divided into four groups:control group,ANP group,LY294002 group and ANP combined LY294002 group.CCK8 was used to detect cell proliferation,scratch test was used to detect cell migration,cell morphology was observed under an inverted microscope and Western blot was used to detect the expression of key signal molecules total Akt(t-Akt)and p-Akt protein of PI3K/Akt signaling pathway.Results:CCK8 results showed that compared with control group,ANP group and LY294002 group,the combination of ANP and LY294002 can inhibit the proliferation of MGC-803 cells,with statistically significant differences(P<0.05);the inhibition of ANP alone and LY294002 alone on the proliferation of MGC-803 cells was time-dependent and dose-dependent.The 24-hour scratch area repair rate of the control group,ANP group,LY294002 group and ANP combined LY294002 group were(50.99±1.80)%,(43.33±2.22)%,(35.81±4.09)%,(26.67±1.69)% and the difference was statistically significant(P<0.05).Four groups of MGC-803 cells showed different cell morphology changes.Both ANP and LY294002 decreased the expression level of p-Akt protein in MGC-803 cells,and ANP combined with LY294002 had a more significant inhibitory effect on the expression of p-Akt protein than they were used alone(P<0.05).The expression level of t-Akt had no significant change in the four groups of cells(P>0.05).Conclusion:ANP can inhibit the proliferation and migration of human gastric cancer cell MGC-803.The combination of ANP and LY294002 can increase the inhibitory effect on gastric cancer cell MGC-803.The me
