摘要
GT6基因属于Trihelix转录因子家族,该家族在调节植物生长发育及应答逆境胁迫等方面发挥着重要作用。为了明确紫花苜蓿Trihelix家族成员的基本特征及其在逆境胁迫下的响应,本研究利用生物信息学方法从蒺藜苜蓿数据库中鉴定出41个Trihelix成员,并对其进行染色体定位、系统进化树和保守基序分析。结果表明,Trihelix家族成员不均匀分布在8条染色体上,其中MtGT6定位在1号染色体上,其分子量为64.373 kD,等电点为5.895,属于亲水性蛋白;亚细胞定位预测显示该基因位于细胞核;对MtGT6启动子顺式作用元件分析,发现启动子区域存在多种顺式作用元件,推测其能在多种逆境中响应;qRT-PCR分析表明,该基因对盐碱响应更为明显。
The GT6 gene belongs to the family of Trihelix transcription factors,which play an important role in regulating plant growth and development and responding to stress.In order to clarify the basic characteristics of the members in the Trihelix family and their responses to stress,this study identified 41 Trihelix members from the Medicago truncatula database and analyzed chromosomal localization,phylogenetic tree and conserved motifs analysis by bioinformatical mehtods.The results showed that the members of the Trihelix family were unevenly distributed on 8 chromosomes,of which MtGT6 was distributed on the 1 thchromosome,with a molecular weight of64.373 kD and isoelectric point of 5.895,belonging to hydrophilic protein.Subcellular localization prediction indicated that the gene was located in the nucleus.By analyzing the cis-acting elements of MtGT6 promoter,it was found that there were many cis-acting elements in the promoter region,and it was speculated that the cis-acting elements could respond to various adversities.It was speculated that it can respond to stress.qRT-PCR analysis displayed that the gene was more responsive to salt and alkali.
