壶瓶碎米荠粗黄酮提取工艺优化及硒对粗黄酮含量和成分的影响 被引量:5

摘要 【目的】探索壶瓶碎米荠中粗黄酮提取的最佳工艺、硒对壶瓶碎米荠粗黄酮提取率的影响以及硒对粗黄酮成分的影响.【方法】通过单因素试验法和正交试验法探索乙醇体积分数、提取温度和时间以及料液比对粗黄酮提取率的影响,采用微波消解-原子荧光法检测硒含量,液-质联用法测定黄酮成分变化.【结果】壶瓶碎米荠粗黄酮提取最佳工艺为乙醇体积分数45%、提取温度65℃、提取时间5 h、料液比为1∶20,在此条件下粗黄酮提取率达到1.9025%.不同质量浓度硒处理下的壶瓶碎米荠粗黄酮提取率不同,硒质量浓度为0~50μg/mL时黄酮提取率呈上升趋势,大于50μg/mL时,粗黄酮提取率呈下降趋势,但粗黄酮中硒含量呈上升趋势.主效成分分析(PCA)结果表明,高质量浓度硒处理壶瓶碎米荠对其叶和根黄酮类物质有显著影响.根中五甲氧基黄酮含量呈上升的趋势,单宁和5,7-二羟基-2-苯并吡喃-4-酮5-O-己糖苷衍生物的含量呈下降趋势.叶中木犀草素C-sinapoyl己糖苷的含量呈上升的趋势,单宁、五甲氧基黄酮等6种黄酮类物质含量呈下降趋势.【结论】研究结果为壶瓶碎米荠粗黄酮提取以及进一步深入研究提供依据. 【Objective】In order to explore the optimum extraction process of crude flavonoids in Cardamine hupingshanensis,the effect of selenium on the extraction rate and components of selenium-containing crude flavonoids in C.hupingshanensis.【Method】The single factor and orthogonal experiments was used to screen optimum factors.The content of selenium of crude flavonoids was detected by double channel atomic fluorescence spectrometry after the sample had been digested by microwave,changes in flavonoid composition were measured by liquid chromatography-mass spectrometry.【Result】The extraction rate of crude flavonoids reached at 1.9025%when ethanol concentration at 45%,and ratio of material to liquid 1∶20,for 5 hours at 65℃.The extraction rate of flavonoid increased with the concentrations of selenium added into nutrient solution at range of 0~50μg/mL,the opposite change occurred as more than 50μg/mL.The selenium content of crude flavonoids increased.The result of principle components analysis showed that treatment with the high concentration selenium had a significant influence on the flavonoid of leaf and root in C.hupingshanensis.the content of pentamethoxy flavone in the roots increased and the contents of tannin and 5,7-dihydroxy-2-benzopyran-4-one 5-O-hexoside derivative reduced.The content of luteolin C-sinapoyl hexoside in the leaves was increased and the content of six flavonoids such as tannin and pentamethoxy flavone decreased.【Conclusion】The results provide the basis for the extraction and further study of crude flavonoids in C.hupingshanensis.
