摘要
目的研究扶正抗癌方的毒理学反应。方法取40只SPF级雌雄各半ICR小鼠,按性别随机分为对照组和扶正抗癌方低、中、高剂量组,每组10只。每天9:00和15:00扶正抗癌方低、中、高剂量组分别给予1.156 g/mL、2.312 g/mL、4.624 g/mL扶正抗癌方灌胃,每次2 mL/100 g,对照组灌胃相同体积的生理盐水,均连续灌胃14 d。观察各组小鼠的一般情况、中毒情况,检测各组小鼠血常规[白细胞(WBC)、红细胞(RBC)、血小板(Plt)计数及血红蛋白(Hb)]、肝功能指标[谷丙转氨酶(ALT)、谷草转氨酶(AST)、总蛋白(TP)、白蛋白(ALB)、碱性磷酸酶(ALP)]、肾功能指标[尿素氮(BUN)、血肌酐(Cr)、尿酸(UA)]水平,记录各组小鼠心、肝、脾、肺、肾的脏器指数,并观察各脏器的病理组织结构。结果实验过程中,各组小鼠的皮毛光滑,饮食量正常,行为及活动正常自如,大小便未见异常,体质量均呈逐渐增加趋势,且各组同一时间点体质量比较差异均无统计学意义(P均>0.05)。各组均未出现中毒相关表现及死亡情况。扶正抗癌方低、中、高剂量组小鼠WBC、Hb呈逐渐升高趋势,Plt呈逐渐下降趋势,但各组间比较差异均无统计学意义(P均>0.05)。各组小鼠同一肝功能和肾功能指标水平、同一脏器指数比较差异均无统计学意义(P均>0.05);各脏器均未见组织增生、炎症及萎缩等病理改变。结论不同剂量扶正抗癌方均无明显的急性毒理学反应,该中药复方可安全用于临床。
Objective It is to study the toxicological reaction of Fuzheng Kangai Decoction.Methods 40 SPF-grade half male and half female ICR mice were randomly divided into control group and low,medium and high dose groups of Fuzheng Kangai Decoction,10 mice in each group.Every day at 9:00 and 15:00,the low,medium and high dose groups of Fuzheng Kangai Decoction were respectively given 1.156 g/mL,2.312 g/mL,4.624 g/mL Fuzheng Kangai Decoction by gavage,2 mL/100 g each time,the control group was intragastrically administered with the same volume of normal saline,both groups were treated for 14 consecutive days.The general condition and poisoning of the mice in each group were observed,and the levels of the blood routine[white blood cell(WBC),red blood cell(RBC),platelet(Plt)count and hemoglobin(Hb)],liver function index[alanine aminotransferase(ALT),aspartate aminotransferase(AST),total protein(TP),albumin(ALB),alkaline phosphatase(ALP)],renal function indexes[urea nitrogen(BUN),blood creatinine(Cr),uric acid(UA)]were detected,the organ index of the heart,liver,spleen,lung,and kidney of the mice in each group were recorded,and the pathological structure of each organ were observed.Results During the experiment,the mice in each group had smooth fur,normal diet,normal behaviors and activities,no abnormalities in urination and defecation,and their body mass had a gradual increase,there was no statistical significant difference in body mass between the groups at the same time point(all P>0.05).No poisoning-related manifestations and deaths occurred in each group.The WBC and Hb of mice in the low,medium and high dose groups of Fuzheng Kangai Decoction showed a gradual increase,and Plt showed a gradual decrease,but there was no statistically significant difference between thee two groups(all P>0.05).There was no significant difference in the same liver and kidney function index levels and the same organ index of the mice in each group(all P>0.05);there were no pathological changes such as tissue proliferation,inflammation and atr
