摘要
目的研究丙泊酚处理对链脲菌素诱导糖尿病大鼠外周血microRNA-126(miR-126)的影响。方法SD雄性大鼠36只适应性饲养1周后,随机数字表法分为正常对照组(n=8)和模型组(n=28)。正常对照组大鼠用普通饲料饲养,模型组大鼠用高糖高脂饲料饲养。链脲菌素诱导的糖尿病大鼠造模成功后随机分配到模型对照组(n=8,给予等体积对照溶剂)和丙泊酚低(n=9,给予低剂量的丙泊酚)、高剂量组(n=8)(给予高剂量的丙泊酚);正常对照组大鼠给予等体积对照溶剂。药物或溶剂输注结束时检测外周血超氧化物歧化酶(SOD)活性,一氧化氮(NO)、可溶性血管细胞粘附分子1(sVCAM-1)和miR-126含量,主动脉miR-126含量。对外周血miR-126与SOD活性、NO、sVCAM-1、主动脉miR-126进行Pearson相关性分析。结果相比正常对照组大鼠,模型对照组大鼠的SOD活性、外周血miR-126和主动脉miR-126水平均显著降低,NO和sVACM-1水平均显著升高,差异有统计学意义(P<0.05)。与模型对照组比较,丙泊酚剂量组SOD活性、外周血miR-126、主动脉miR-126水平显著升高,外周血NO、sVCAM-1水平显著降低,并呈剂量依赖性,差异有统计学意义(P<0.05)。外周血miR-126与SOD活性和主动脉miR-126呈正相关(r=0.845,0.878,P<0.05),与外周血NO和sVACM-1呈负相关(r=-0.922,-0.925,P<0.05)。结论丙泊酚对糖尿病大鼠血管内皮细胞损伤有一定保护作用,并能显著升高STZ诱导糖尿病大鼠外周血miR-126水平。miR-126与糖尿病血管内皮细胞功能生物标记物可能有相关性。
Objective To study the effect of propofol treatment on microRNA-126 in peripheral blood in the streptozotocin-induced rat model with diabetes mellitus.Methods After 36 male SD rats were adaptively reared for 1 week,they were randomly divided into normal control group(n=8)and model group(n=28).Normal control rats were fed with ordinary feed,and model rats were fed with high-sugar and high-fat feed.After the rat model with diabetes mellitus induced by streptozotocin was made successfully,they were randomly assigned to the model control group(n=8,given equal volume of control solvent)and propofol low(n=9,given low dose of propofol),high Dose group(n=8)(given high-dose propofol);rats in the normal control group were given an equal volume of control solvent.At the end of infusion,the activity of superoxide dismutase(SOD),the levels of nitric oxide(NO),soluble vascular cell adhesion molecule1(sVCAM-1)and miR-126 in peripheral blood,and the level of miR-126 in aorta were measured.Peripheral blood miR-126 and SOD activity,NO,sVCAM-1,aortic miR-126 content were analyzed by Pearson correlation.Results Compared with those in normal rats,in the rats with diabetes mellitus,the activity of SOD,the level of miR-126 in peripheral blood and in aorta all decreased significantly,and the level of NO and sVACM-1 in peripheral blood all increased(P<0.05).Propofol increased the activity of SOD,the level of miR-126 in peripheral blood and in aorta,and decreased the level of NO and sVACM-1 in peripheral blood dose-dependently(P<0.05).The level of miR-126 in peripheral blood was positively correlated with the activity of SOD and the level of miR-126 in aorta(r=0.845,0.878,P<0.05);while it was negatively correlated with the level of NO and sVACM-1 in peripheral blood(r=-0.922,-0.925,P<0.05).Conclusion Propofol has a protective effect on vascular endothelial cell injury in diabetic rats,and can significantly increase the levels of miR-126 in the peripheral blood of the STZ-induced rat model with diabetes mellitus;miR-126 may be related to b
