子宫腺肌病子宫内膜-肌层交界区平滑肌细胞中Lin28表达及对细胞增殖影响研究 被引量:6

Expression of Lin28 in the smooth muscle cells of endometrial-myometrial interface of human adenomyosis and its effect on cell proliferation

摘要 目的探讨Lin28在子宫腺肌病(ADS)子宫内膜-肌层交界区(EMI)平滑肌细胞中的表达及对细胞增殖功能的影响。方法收集2018年2月至2019年5月于首都医科大学附属北京妇产医院因ADS行子宫切除术的患者16例为ADS组;收集同期因宫颈上皮内瘤变3级、子宫颈癌ⅠA期以及卵巢上皮性肿瘤行子宫切除术的患者12例为对照组。对两组患者EMI平滑肌细胞进行分离及原代培养。采用qRT-PCR、Western-blot检测两组EMI平滑肌细胞中Lin28A和Lin28B mRNA及蛋白的表达水平;Lin28B特异siRNA转染ADS中EMI平滑肌细胞,用CCK8法检测Lin28B表达沉默对细胞增殖能力的影响。结果(1)Lin28B在ADS组EMI平滑肌细胞中mRNA和蛋白表达水平均高于对照组,差异有统计学意义(P<0.01);两组细胞中Lin28A mRNA和蛋白表达水平差异无统计学意义(P>0.05)。(2)ADS患者EMI平滑肌细胞增殖速度较对照组细胞快,转染Lin28B siRNA后显著抑制细胞生长,转染后48、60、72及84h细胞生长数量(即吸光度值,OD值)依次为0.86±0.11、1.29±0.15、1.69±0.18、2.05±0.08,空白对照组细胞的OD值依次为1.08±0.13、1.61±0.07、2.29±0.12、2.61±0.07,分别比较差异均有统计学意义(P<0.01)。结论Lin28B在ADS子宫EMI平滑肌细胞中呈高表达,可能导致EMI平滑肌细胞增殖能力亢进,从而参与ADS发生发展。 Objective To observe the expression of Lin28 in the smooth muscle cell of endometrial-myometrial interface(EMI)of adenomyosis(ADS)and the effect on cell proliferation.Methods From Feb.2018 to May 2019,16 cases withADS undergoing hysterectomy were enrolled as ADS group,matched with 12 cases of hysterectomy due to cervicalintraepithelial neoplasia grade 3,cervical carcinoma stageⅠA and epithelial ovarian carcinoma as control group inBeijing Obstetrics and Gynecology Hospital Affiliated to Capital Medical University.EMI smooth muscle cells wereisolated and cultured immediately after the operation.The expression of m RNA and protein of Lin28 A and Lin28 B inEMI smooth muscle cells in two groups were measured with q RT-PCR and Western-blot;Lin28 B specific si RNA wastransfected into EMI smooth muscle cells of ADS.The effect of Lin28 B-deficiency on the proliferation of smooth musclecells was detected by CCK8.Results(1)The expression levels of m RNA and protein of Lin28 B in EMI smooth musclecells of ADS group were significantly higher than those of the control group(P<0.01).The expression level of proteinand mRNA of Lin28 A showed no significant difference between two groups(P>0.05).(2)The proliferation rate of EMI smooth muscle cells of ADS was more intensive than thatof control group.After transfecting Lin28 B si RNA,thecell growth was significantly inhibited and at 48,60,72 and 84 hours after transfection with Lin28 B si RNA,thenumber of cell growth(absorbance value,OD value)inturn was 0.86±0.11,1.29±0.15,1.69±0.18 and 2.05±0.08,respectively;the OD value in control group was 1.08±0.13,1.61±0.07,2.29±0.12 and 2.61±0.07,respectively.The differences being statistically significant(P<0.01).Conclusion The expression of Lin28 B in EMI smooth muscle cells of ADS is significantly increased,which may leadto abnormal proliferation of EMI smooth muscle cells.Thus Lin28 B may be involved in the development of ADS.
