摘要
目的探讨结直肠癌中错配修复基因MLH1、MSH2、MSH6和PMS2与Ang-1、Ang-2的表达及相关性。方法收集2015年1月~2018年12月北华大学附属医院病理科结直肠癌石蜡标本,所有病例术前均未进行放、化疗。利用免疫组化Envision法检测MLH1、MSH2、MSH6、PMS2、Ang-1和Ang-2蛋白的表达,MLH1、MSH2、MSH6和PMS2为细胞核染色,Ang-1、Ang-2为细胞浆染色,阳性结果采用半定量评分标准。采用SPSS 20.0统计学软件,计数资料用频数和百分比表示,利用χ2检验、Fisher确切概率法分析MLH1、MSH2、MSH6、PMS2、Ang-1、Ang-2与临床病理特征的关系,利用Spearman相关法分析MLH1、MSH2、MSH6、PMS2与Ang-1、Ang-2的相关性,P<0.05为差异有统计学意义。结果(1)MLH1、PMS2缺失与患者的性别、年龄、淋巴结转移、大体类型、组织学类型比较,差异无统计学意义(P>0.05),与发病部位、分化程度比较,差异有统计学意义(P<0.05);MSH2、MSH6缺失与患者的性别、年龄、发病部位、分化程度、淋巴结转移、大体类型、组织学类型比较,差异无统计学意义(P>0.05)。(2)MLH1与PMS2之间显著正相关,MSH2与MSH6之间显著正相关且协同表达。(3)Ang-1、Ang-2表达与患者的性别、年龄、分化程度、发病部位、大体类型、组织学类型比较,差异无统计学意义(P>0.05),但与淋巴结转移比较,差异有统计学意义(P<0.05)。(4)MLH1、MSH2、MSH6和PMS2缺失与Ang-1、Ang-2的表达比较,差异无统计学意义(P>0.05)。结论MLH1、MSH2、MSH6、PMS2缺失与Ang-1、Ang-2表达无显著相关性。
Objective To investigate the correlation between mismatch repair genes of MLH1,MSH2,MSH6 and PMS2 and Ang-1/2 expressions in colorectal cancer.Methods Paraffin embedded specimens of patients with colorectal cancer from January 2015 to December 2018 were collected from the Pathology Department of Affiliated Hospital of Beihua University.All cases were not treated with radiotherapy and chemotherapy before operation.Immunohistochemistry Envision method was used to detect the protein expressions of MLH1,MSH2,MSH6,PMS2,Ang-1 and Ang-2,in which MLH1,MSH2,MSH6 and PMS2 received nuclear staining,and Ang-1 and Ang-2 received cytoplasmic staining.Meanwhile,semi-quantitative scoring standard was used for positive results.Counting data expressed in frequency and percentage,SPSS20.0 statistical software was used to analyze the correlation between MLH1,MSH2,MSH6,PMS2,Ang-1,Ang-2 and the clinicopathological features by chi square test and Fisher exact probability method.Meanwhile,the correlation between MLH1,MSH2,MSH6,PMS2 and Ang-1,Ang-2 was analyzed by Spearman correlation method,and P<0.05 was considered statistically significant.Results(1)There was no statistical difference between the absences of MLH1 and PMS2 and the gender,age,lymph node metastasis,gross morphology and histological type of the patients(P>0.05).While there was statistical difference between it and the site of pathological changes and differentiation degree(P<0.05).There were no statistical differences between the absences of MSH2 and MSH6 and the gender,age,site of pathological changes,differentiation degree,lymph node metastasis,gross morphology and histological type of patients(P>0.05).(2)There were significantly positive correlation between MLH1 and PMS2,and significantly positive correlation and synergistic expression between MSH2 and MSH6.(3)There were no statistical differences between the expressions of Ang-1 and Ang-2 and the gender,age,differentiation degree,site of pathological changes,gross morphology and histological type of patients(P>0.05).W
