响应面法优化黔产大宗药材苦参中总生物碱的提取工艺研究

摘要 苦参是中国的传统中药之一,临床主要用于治疗痢疾、便血、黄疸、湿疹及妇科炎症等病症,其中生物碱是其主要药效成分.主要是采用乙醇回流提取法来研究考察黔产苦参中总生物碱的最佳工艺.以苦参总生物碱的提取率作为考察指标,采用乙醇回流提取法,考察提取温度、提取次数、提取时间、料液比以及乙醇浓度对总生物碱的影响;接着在单因素试验结果基础上,采用计算机软件Box-Benhnken实验设计方法优化最佳提取工艺条件.结果显示,其总生物碱的最佳工艺条件为:乙醇浓度85%,提取温度80℃,提取时间2 h,提取次数3次,料液比1∶9;并且在最佳提取工艺条件的验证性试验中,黔产大宗药材苦参总生物碱的提取率达到45.3%,这说明了本试验的提取率优于其它提取工艺方法.该研究方法操作简易及实验设备简单,可为黔产苦参用于工业化提取总生物碱提供一定的理论依据. Sophora flavescens is a popular traditional Chinese herbal medicine in China.It shows significant pharmacological activities in the treatment of dysentery,hematochezia,jaundice,eczema and gynecological inflammation.The optimum process for total alkaloids from dried roots of S.flavescens from Guizhou province was investigated through ethanol reflux method.The total alkaloids’extraction ratio was used as the index,and extraction temperature,times,material-liquid ratio and ethanol concentration were single factor.Furthermore,the best extraction process was built by computer software Box-Benhnken design method based on the single index experiment.The result showed that the way to obtain optimal conditions was ethanol concentration 85%,temperature 80℃,extracting time 2 hours,extraction times 3 times and material-liquid ratio 1:9.And the extraction rate of total alkaloids was up to 45.3%under these conditions.The research method is easy to operate and experimental equipment is simple,which will contribute to the development of extraction process of S.flavescens as medicines in the industrial application.
