RP-HPLC法测定回生口服液中3种成分的含量

摘要 目的建立RP-HPLC法同时测定回生口服液中山柰酚、阿魏酸和迷迭香酸3种活性成分的含量。方法采用Shim-pack CLC-ODS C_(18)色谱柱分离;检测波长:342 nm;柱温:32℃;进样体积:20μL;流动相:乙腈(A)-1 mL·L^(-1)甲酸(B);梯度洗脱,洗脱程序为:0~5 min,11%A;5~18 min,11%A^35%A;18~26 min,35%A^69%A;26~28 min,69%A^11%A;29~35 min,11%A。外标法定量检测。结果 3种待测组分质量浓度均在0.1~20.0μg·mL^(-1)范围内线性关系良好;重复性和精密度RSD值均小于5.0%(n=6);平均回收率分别为101.12%,98.26%和98.21%;供试品溶液在24 h内稳定性良好。结论此方法简单、快速,结果准确,重复性、精密度和稳定性均较好,可用于回生口服液中中成药产品的质量控制。 Objective To develop a reverse phase high-performance liquid chromatography(RP-HPLC)method for simultaneously determining the contents of kaempferol,ferulic acid and rosmarinic acid in Huisheng Oral Liquid.Methods Shim-pack CLC-ODS C 18 column was selected for separation,and detection wavelength was set at 342 nm.The column temperature was 32℃,and the injection volume was 20μL.The mobile phase was acetonitrile(A)and 1 mL·L-1 formic acid(B).Gradient elution procedure was as follows:0-5 min,11%A;5-18 min,11%A-35%A;18-26 min,35%A-69%A;26-28 min,69%A-11%A;29-35 min,11%A.External standard method was used for quantitative detection.Results The mass concentrations of the 3 components showed good linear relationships in the range of 0.1 to 20.0μg·mL-1.The RSD of repeatability and precision were all below 5.0%(n=6).The average recoveries of the 3 active components were 101.12%,98.26%and 98.21%,respectively.The sample solution was stable within 24 h.Conclusion This method is simple,fast,accurate,reproducible,precise and stable,and can be used for the quality control of Chinese patent medicine Huisheng Oral Liquid.
