毛排钱草的体外抑菌活性及稳定性评价 被引量:4

摘要 目的:考察毛排钱草的体外抑菌活性及其抑菌稳定性。方法:采用牛津杯法、微量肉汤稀释法考察毛排钱草根、茎和叶95%乙醇提物的抑菌活性,明确活性部位;以金黄色葡萄球菌和铜绿假单胞菌为指示菌,探讨热处理、紫外线、介质pH和常见金属离子对毛排钱草提取物抑菌稳定性的影响。结果:毛排钱草根、茎和叶在实验浓度范围内均具有较好的抑菌效果,其作用强弱为根>叶>茎;其中根的正丁醇层对金黄色葡萄球菌的抑菌作用较为显著,抑菌圈直径为(24.04±0.78)mm,最低抑菌浓度(minimum inhibitory concentration,MIC)和最低杀菌浓度(minimum bactericidal concentration,MBC)均为15.63 mg·mL^-1生药浓度。紫外照射10 min、60~121℃加热处理、偏酸性环境(pH<5.9)及Na^+、K^+、Ca^2+、Fe^3+等金属离子的存在对于提取物的抑菌活性均产生了抑制作用,其抑制程度与敏感菌存在一定的关联性。结论:毛排钱草根、茎和叶均具有良好的体外抑菌活性,在实验设定的不同条件下表现出较好的抑菌稳定性,具有进一步开发和利用的前景。 Objective:To investigate in vitro antibacterial activity of Phyllodium elegans(Lour.)Desv.and its antibacterial stability.Methods:The antibacterial activity of 95%ethanol extracts from the roots,stems,and leaves of P.elegans was investigated by the Oxford cup and the broth dilution.Staphylococcus aureus and Pseudomonas aeruginosa were used as indicator bacteria to investigate the effects of heat treatment,ultraviolet rays,pH of the medium and common metal ions on the antibacterial stability of the extract of P.elegans.Results:The roots,stems,and leaves of P.elegans had good bacteriostatic effects in the experimental concentration range,and the effects were as follows:roots>leaf>stems.The n-butanol layer of the root has a significant antibacterial effect on Staphylococcus aureus.The diameter of the inhibition zone is(24.04±0.78)mm,the minimum inhibitory concentration(MIC)and the minimum bactericidal concentration(MBC)are all 15.63 mg·mL^-1.Ultraviolet irradiation for 10 min,heat treatment at 60-121℃,acidic environment(pH<5.9),metal ions such as Na^+,K^+,Ca^2+,Fe^3+ were all have an inhibitory effect on the bacteriostatic activity of the extract,and there was correlation between Staphylococcus aureus and Pseudomonas aeruginosa.Conclusion:The roots,stems,and leaves of P.elegans have good antibacterial activity in vitro,and show good antibacterial stability under different conditions in experiment,which show prospect of development and utilization.
