摘要 目的建立UPLC-MS/MS法测定人血浆中5种精神类药物浓度。方法样品经乙腈沉淀蛋白处理,以奥卡西平为内标,用Waters XBridge BEH C18色谱柱(100 mm×2.1 mm,2.5μm),以0.1%甲酸水-乙腈为流动相,梯度洗脱,流速为0.3 mL·min^-1。用电喷雾离子源,正离子模式检测,MRM模式扫描。结果氯普噻吨、氟奋乃静、奋乃静、硫利达嗪和异丙嗪线性范围均为10~500 ng·mL^-1,相关系数大于0.9960。定量下限为10 ng·mL^-1;日内、日间RSD均小于15%。结论 UPLC-MS/MS方法快速、灵敏、准确,适用于人血浆中氯普噻吨、氟奋乃静、奋乃静、硫利达嗪和异丙嗪的同时检测。 Objective To develop a method for quantification of five traditional antipsychotics in human plasma. Methods Sample was treated with acetonitrile. Oxcarbazepine was used as internal standards. The detection was performed on Waters XBridge BEH C18 column(100 mm×2.1 mm, 2.5 μm) by the gradient elution with 0.1% formic acid water and acetonitrile as mobile phase, at 0.3 mL·min^-1 flow rate. ESI was used for positive ion scanning by multiple reaction monitoring mode. Results The linear range of chlorprothixene,perphenazine,fluphenazine,thioridazine and promethazine ranged from 10 ng·mL^-1 to 500 ng·mL^-1(r>0.9960).The limit of quantitation were 10 ng·mL^-1. The RSD of inter and intra-day were less than 15%. Conclusion The method is rapid, sensitive and accurate, which can be used for simultaneous quantification of chlorprothixene,perphenazine, fluphenazine, thioridazine and promethazine in human plasma.
