摘要
目的研究黄芪总皂苷对脑出血大鼠脑组织病理形态学及水通道蛋白-4(AQP-4)、水通道蛋白-9(AQP-9)、基质金属蛋白酶-9(MMP-9)表达的影响。方法采用改良的自体动脉血法制备脑出血大鼠模型,将建模成功的脑出血大鼠随机分为模型组(n=20)与实验组(n=20);假手术组(n=20)大鼠仅裸露颈总动脉、神经,不注射自体动脉血。实验组大鼠腹腔注射200 mg·kg-1·d-1黄芪总皂苷,假手术组与模型组腹腔注射等量生理盐水,每天1次,连续干预7 d。用干-湿重法测定各组大鼠的脑组织含水量;用苏木精-伊红(HE)染色观察脑组织病理形态学变化;用蛋白质印迹(Wb)法检测脑组织AQP-4、AQP-9、MMP-9蛋白的表达水平;用酶联免疫吸附(ELISA)法检测脑组织过氧化氢酶(CAT)、超氧化物歧化酶(SOD)活性与丙二醛(MDA)含量。结果假手术组、模型组与实验组大鼠脑组织含水量分别为(57.84±2.24)%,(94.46±3.10)%,(69.87±1.93)%;脑组织AQP-4蛋白相对表达量分别为0.21±0.09,0.82±0.13,0.47±0.11;AQP-9蛋白相对表达量分别为0.18±0.06,0.94±0.06,0.41±0.10;MMP-9蛋白相对表达量分别为0.08±0.03,0.80±0.12,0.59±0.14。与假手术组比较,模型组大鼠脑组织含水量、MDA含量及AQP-4、AQP-9、MMP-9蛋白相对表达量均明显升高,而实验组低于模型组;模型组大鼠脑组织SOD与CAT活性均显著低于假手术组,而实验组高于模型组(均P<0.05)。结论黄芪总皂苷可显著改善脑出血大鼠脑组织病理学形态,缓解脑水肿,同时降低氧化应激损伤,保护脑组织,其作用机制可能与降低脑出血大鼠脑组织AQP-4、AQP-9、MMP-9蛋白表达相关。
Objective To investigate the effect of total saponins of astragalus on pathological morphology of brain tissues and expression of aquaporin-4(AQP-4), aquaporin-9(AQP-9) and matrix metalloproteinase-9(MMP-9) of rats with cerebral hemorrhage. Methods The rat model of cerebral hemorrhage were established by a modified autologous arterial blood method, the successfully established cerebral hemorrhage rats were randomly divided into model group(n=20) and test group(n=20);the rats in sham-operation group(n=20) were only exposed the common carotid artery and nerve, and no autologous arterial blood was injected. The rats in test group were intraperitoneally injected with astragalus saponins 200 mg·kg-1·d-1,sham-operation group and model group were intraperitoneally injected with the same amount of normal saline,1 time·d-1,and for 7 d. The dry-wet weight method was used to determine the water content of the brain tissues in each group of rats;hematoxylin-eosin(HE) staining was used to observe the pathological morphological changes of brain tissues;Western blotting(Wb) method was used to detect the expression levels of AQP-4,AQP-9 and MMP-9 protein in brain tissues;enzyme linked immunosorbent assay(ELISA) was used to detect the activity of catalase(CAT),superoxide dismutase(SOD) and malondialdehyde(MDA) in brain tissues. Results The water content of brain tissues in sham-operation group,model group and test group were(57. 84 ± 2. 24) %,(94. 46 ± 3. 10) %,(69. 87 ± 1. 93) %;the relative expression levels of AQP-4 protein in brain tissues were 0. 21 ± 0. 09,0. 82 ± 0. 13,0. 47 ± 0. 11;the relative expression levels of AQP-9 protein were 0. 18 ± 0. 06,0. 94 ± 0. 06,0. 41 ± 0. 10;the relative expression levels of MMP-9 protein were 0. 08 ± 0. 03,0. 80 ± 0. 12,0. 59 ± 0. 14.
