摘要
建立了高效液相色谱测定小麦粉中脱氧雪腐镰刀菌烯醇的分析方法,比较了直接浸提法、固相萃取柱法和免疫亲和柱法3种前处理方法对测定结果的影响。结果表明:直接浸提法容易受到样品中杂质组分干扰,测定低浓度样品时回收率和精密度较差;固相萃取柱法和免疫亲和柱法能有效减少样品中的干扰杂质,测定结果稳定,精密度高,回收率均大于85%,适用于小麦粉中脱氧雪腐镰刀菌烯醇的测定。
A method for the determination of deoxynivalenol in wheat flour by high performance liquid chromatography(HPLC)was established.The pretreatment of samples had a great influence on the test results.The effects of direct extraction,solid phase extraction(SPE)and immunoaffinity column were compared.The results of pretreatment using direct extraction method were easy to be interfered by the impurities in the samples,and the recoveries and precision of the low concentration sample were poor.The results of pretreatment using solid phase extraction and immunoaffinity column were better.The two methods could effectively reduce the interference of impurities in the samples.The determination results were stable,high precision,and the recoveries of the standards spiked in real samples were above 85%.They were suitable for the determination of deoxynivalenol in wheat flour.
