摘要
目的探究重楼皂苷Ⅵ对人急性髓系白血病THP-1细胞增殖和凋亡的影响及机制。方法采用CCK8实验考察重楼皂苷Ⅵ对THP-1细胞活力的影响,并确定给药剂量。流式细胞术检测重楼皂苷Ⅵ对THP-1细胞凋亡的影响。Western blot进一步检测细胞凋亡与p38 MAPK通路相关蛋白的变化,通过p38抑制剂SB203580验证THP-1细胞中上述通路与凋亡的关系。结果一定剂量的重楼皂苷Ⅵ能抑制THP-1细胞增殖且抑制效应呈浓度依赖与时间依赖性。流式细胞术显示,与对照组比较,给药组细胞凋亡率明显升高(P<0.05)。Western blot进一步表明,随着药物浓度增加,cleaved caspase-3与Bax蛋白表达增多,Bcl-2蛋白表达下降,p-p38/p38比值增加(P<0.05),上述作用可被p38 MAPK抑制剂SB203580阻断。CCK8实验结果显示,抑制p38通路可降低药物导致的细胞增殖抑制。结论重楼皂苷Ⅵ可抑制急性髓系白血病THP-1细胞增殖并通过p38 MAPK通路诱导细胞凋亡。
This study was performed to investigate the effect of polyphyllin Ⅵ(PPⅥ) on human acute myeloid leukemia THP-1 cells and the mechanism. CCK8 assay was used to investigate the effect of PPⅥ on the activity of THP-1 cells;flow cytometry was used to detect the effect of PPⅥ on the apoptosis of THP-1 cells;Western blot was used to further detect the changes of proteins related to apoptosis and p38 MAPK pathway, and p38 MAPK inhibitor SB203580 was introduced to verify the relationship between the above pathway and apoptosis in THP-1 cells. Data showed that a certain dose of PP Ⅵ could inhibit the proliferation of THP-1 cells, and the inhibitory effect was concentration dependent and time dependent. Flow cytometry showed that the apoptosis rate was significantly increased in the administration group compared with the control group. Western blot further showed that, as the drug concentration increased, the protein expression of cleaved caspase-3 and Bax increased, while Bcl-2 protein expression decreased, the ratio of p-p38/p38 was increased. The above effect could be blocked by the p38 MAPK inhibitor SB203580. CCK8 assay showed that inhibition of p38 pathway could reduce the inhibition of cell proliferation induced by PP Ⅵ. Taken together, PPⅥcould significantly inhibit the proliferation of acute myeloid leukemia THP-1 cells and induce apoptosis through the p38 MAPK pathway.
