沈阳花生上菜豆普通花叶病毒的鉴定与全序列分析

摘要 2017年9月在辽宁沈阳采集到表现明显斑驳褪绿症状的花生样品,怀疑为植物病毒侵染所致.透射电镜负染色观察到约15 nm×800 nm的线性病毒粒子.选取明显病症样品进行转录组测序,原始数据经过滤拼接后进行同源搜索,显示该病毒与菜豆普通花叶病毒(Bean common mosaic virus,BCMV)高度同源.基于转录组结果,设计6对特异引物扩增BCMV全基因组,确定其全长为10076bp核苷酸(MN786956).基于外壳蛋白基因(coat protein,CP)进行同源性比对,结果显示,其与BCMV中国武汉大豆分离物(BCMV-[CN:WH:Glycine max],KJ807813)的核苷酸同源性为99.4%,氨基酸同源性为99.7%.系统发育分析显示它聚类在BCMV花生分离物的分支上.这是辽宁地区有关BCMV自然侵染花生的首次报道. In September 2017,peanut samples with obvious mottled chlorosis symptoms were collected in Shenyang,Liaoning Province,suspected to be infected by plant viruses.Negative staining of transmission electron microscope observed linear virus particles of about 15 nm×800 nm.Samples with obvious diseased symptoms were selected for transcriptome sequencing.The original data were filtered and spliced for homology search,which showed a high degree of homology with Bean common mosaic virus(BCMV).Based on the transcriptome results,six pairs of specific primers were designed to amplify the entire genome of BCMV,and its full length was determined to be 10076 bp nucleotides(MN786956).Based on the homology analysis of CP,it was found that it has the highest homology with BCMV Wuhan soybean isolate(BCMV-[CN∶WH∶Glycine max],KJ807813),with nucleotide homology of 99.4%and amino acid homology of 99.7%.Phylogenetic analysis showed that it clustered on the branch of BCMV peanut isolate.Therefore,it is believed that it is BCMV that infects peanuts.This is the first report of BCMV infecting peanuts naturally in Liaoning.
