摘要
目的探讨丹皮酚对大鼠心肌缺血再灌注损伤的保护作用及对Toll样受体4(TLR4)、白细胞介素-23(IL-23)及超氧化物歧化酶(SOD)活性的影响。方法通过结扎大鼠左冠脉前降支30 min再灌注3 h建立大鼠心肌缺血再灌注损伤模型,将SD大鼠经随机方式划分成4组,各组均为13只SD大鼠,分为假手术组、模型组、丹皮酚低剂量组(15 mg/kg)、丹皮酚高剂量组(30 mg/kg),于术前10 d开始灌胃给药。再灌注3 h后取血清,采用比色法测定大鼠心肌SOD活性,通过2,3,5-氯化三苯基四氮唑试剂染色法测定大鼠心肌梗死面积,免疫组化检测TLR4阳性细胞平均积分光密度(AIOD),蛋白质免疫印迹法测定大鼠心肌组织IL-23蛋白水平。结果模型组、丹皮酚低剂量组、丹皮酚高剂量组大鼠心肌组织IL-23蛋白水平、TLR4阳性细胞AIOD、SOD活性与假手术组比较,差异有统计学意义(P<0.05);丹皮酚低剂量组、丹皮酚高剂量组大鼠心肌组织IL-23蛋白、TLR4阳性细胞AIOD、SOD活性与模型组比较,差异有统计学意义(P<0.05)。结论使用丹皮酚预处理可通过降低TLR4炎性作用,减少IL-23释放,增强SOD活性,从而实现对大鼠心肌缺血再灌注损伤的保护效能。
Objective To investigate the protective effect of paeonol on myocardial ischemia-reperfusion injury in rats and its effects on Toll-like receptor 4(TLR4),interleukin-23(IL-23)and superoxide dismutase(SOD)activity.Methods The myocardial ischemia-reperfusion injury model of rats was established by ligating the descending branch of left coronary artery for 30 min and reperfusion for 3 h.SD rats were randomly divided into 4 groups,each consisting of 13 SD rats,which were divided into the sham operation group,the model group,the low-dose paeonol group(15 mg/kg)and the high-dose paeonol group(30 mg/kg).The rats were given orally 10 days before the operation.Serum was collected 3 h after reperfusion,and SOD activity in rat myocardium was determined by colorimetric method.Myocardial infarction area was determined by 2,3,5-triphenyltetrazolium chloride staining method,average integrated optical density of TLR4-positive cells was detected by immunohistochemistry,and IL-23 protein level in rat myocardium was determined by Western blot.Results Compared with the sham operation group,the model group,the low-dose paeonol group and the high-dose paeonol group,the levels of IL-23 protein,TLR4-positive cells AIOD and SOD activity were statistically significant(P<0.05).Compared with the model group,the activities of IL-23 protein,TLR4-positive cells AIOD and SOD activity in the rat myocardium of the low-dose paeonol group and high-dose paeonol group were statistically significant(P<0.05).Conclusion Pretreatment with paeonol can reduce the inflammatory effect of TLR4,reduce the release of IL-23,and enhance the activity of SOD,so as to realize the protective effect of myocardial ischemia reperfusion injury in rats.
