荧光增白剂CXT和CBS的物理化学性质及细胞毒性 被引量:1

摘要 目的研究荧光增白剂CXT和CBS的物理化学性质及其对人支气管上皮细胞16HBE和人皮肤角质细胞HaCaT的生物安全性。方法利用高效液相色谱仪检测紫外吸光度反映荧光增白剂的纯度,利用动态光散射仪检测荧光增白剂分子的尺寸和电荷,使用分光测色仪检测荧光增白剂对棉布的增白效果。以人支气管上皮细胞16HBE和人皮肤角质细胞HaCaT为研究对象,利用酶标仪和MTT试剂检测细胞液在490 nm处的吸光度反映细胞活力,利用显微镜观察细胞膜表面形态变化,利用流式细胞仪检测细胞液荧光强度反映细胞凋亡情况。通过设立空白对照组,50,100,200μg/ml荧光增白剂CXT和CBS组,研究荧光增白剂CBS和CXT对这两种细胞的毒理学效应。结果荧光增白剂CXT和CBS主要成分的纯度在97%左右,对棉布均有良好的增白强度,分别达到99.92%和99.69%,其表面具有负电荷和纳米级的分子尺寸。研究结果显示,与空白对照组的细胞相比,荧光增白剂CBS在100μg/ml时16HBE和HaCaT细胞活力分别降低到80.0%和90.1%,荧光增白剂CXT在100μg/ml时16HBE和HaCaT细胞活力分别降低到88.5%和93.2%,但这两种细胞形态均未发生明显变化。细胞凋亡的流式分析结果也证实,即使在200μg/ml的荧光增白剂溶液作用下,细胞也未发生明显的凋亡。结论荧光增白剂CXT和CBS表面的负电荷使它们具有良好的水溶性,纳米级的分子尺寸使其染色效果更为持久。在日常使用情况下(洗涤剂中的荧光增白剂的添加量为4-40μg/ml),荧光增白剂中无论是CBS还是CXT均不会对呼吸道或者皮肤表面的细胞造成毒性损伤,具有良好的生物安全性。 Objective To study the physicochemical properties of fluorescent whitening agents CXT and CBS and their biological safety to human bronchial epithelial cells 16HBE and human skin keratinocytes HaCaT.Methods High-performance liquid chromatography was used to determine the purity of the fluorescent whitening agent by measuring UV absorbance,and a dynamic light scattering was used to measure the size and charge of the fluorescent whitening agent,and the whitening effect of fluorescent whitening agent on cotton fabric was tested by spectrophotometer.Human bronchial epithelial cell 16HBE and human keratinocyte HaCaT were used as the research objects.The absorption value at 490 nm was detected by enzyme labeled instrument and MTT reagent to evaluate the cell viability,the morphological changes of cell membrane were observed by microscope,and the apoptosis of cells was detected by flow cytometry.The cells were divided into blank control group,50μg/ml,100μg/ml,200μg/ml fluorescent whitening agent CXT and CBS groups.The toxicological effects of fluorescent whitening agents CBS and CXT on 16HBE and HaCaT cells were observed.Results The purity of the main components of the fluorescent brighteners CXT and CBS was about 97%.The whitening strength for cotton cloth was 99.92%and 99.69%,respectively.The fluorescent whitening agents CXT and CBS had negative charge on the surface and nano-level of molecular size.Compared to blank control group,the viabilities of 16HBE and HaCaT cells were decreased to 80.0%and 90.1%in 100μg/ml CBS group,and the viabilities of 16HBE and HaCaT cells were decreased to 88.5%and 93.2%in 100μg/ml CXT group,respectively,but the morphology of these two kinds of cells showed no significant change.The flow cytometric analysis results also confirmed that the cells did not undergo obvious apoptosis even under 200μg/ml fluorescent whitening agent solution.Conclusion The negative charge on the surface makes CXT and CBS have good water solubility.The nano-level of molecular size makes the dyeing effect mor
