摘要 目的:建立HPLC法同时测定鼻咽清毒颗粒中8种活性成分的含量。方法:采用Waters XTERRA C18色谱柱(250mm×4.6 mm,5μm),流动相为乙腈(A)-0.15%磷酸溶液(B),梯度洗脱;检测波长为210 nm(重楼皂苷Ⅰ和重楼皂苷Ⅱ)、254nm(3,5-O-二咖啡酰基奎宁酸)、270 nm(龙胆苦苷)、334 nm(绿原酸、新绿原酸、木犀草苷、蒙花苷),流速:1.0 ml·min^-1,柱温:35℃,进样量:10μl。结果:绿原酸、木犀草苷、蒙花苷、新绿原酸、3,5-O-二咖啡酰基奎宁酸、龙胆苦苷、重楼皂苷Ⅰ和重楼皂苷Ⅱ检测质量浓度的线性范围分别为4.420~44.200μg·ml^-1,2.404~24.040μg·ml^-1,4.868~48.680μg·ml^-1,2.672~26.720μg·ml^-1,0.411~4.112μg·ml^-1,1.038~10.380μg·ml^-1,0.854~8.536μg·ml^-1,1.634~16.340μg·ml^-1(r值范围0.9993~0.9999);平均加样回收率分别为99.78%,98.95%,99.58%,98.87%,98.42%,99.04%,99.27%,99.17%(RSD<1.0%,n=6)。结论:所建立的方法简便、有效、准确,可用于鼻咽清毒颗粒中上述8种活性成分含量的同时测定。 Objective:To establish an HPLC method for the simultaneous determination of eight constituents in Biyan Qingdu granules.Methods:The determination was performed on a Waters XTERRA C18 column(250 mm×4.6 mm,5μm)with gradient elution,and the mobile phase consisted of acetonitrile-0.15%phosphoric acid at the flow rate of 1.0 ml·min^-1.The detection wavelengths were set at 210 nm for aristolochic acidⅠand chonglou saponinⅡ,254 nm for 3,5-O-dicaffeoylquinic acid,270 nm for gentiopicrin,and 334 nm for chlorogenic acid,luteolin-7-glucoside,buddleoside and neochlorogenic acid.The column temperature was set at 35℃and the injection volume was 10μl.Results:The linear range was 4.420-44.20μg·ml^-1 for chlorogenic acid,2.404-24.040μg·ml^-1 for luteolin-7-glucoside,4.868-48.680μg·ml^-1 for buddleoside,2.672-26.710μg·ml^-1 for neochlorogenic acid,0.411-41.120μg·ml^-1 for 3,5-O-dicaffeoylquinic acid,1.038^-10.380μg·ml^-1 for gentiopicrin,0.854-8.536μg·ml^-1 for aristolochic acidⅠand1.634^-16.340μg·ml^-1 for achonglou saponinⅡ(r=0.9993-0.9999).The average recovery was 99.78%,98.95%,99.58%,98.87%,98.42%,99.04%,99.27%and 99.17%(RSD<1.0%,n=6),respectively.Conclusion:The method is simple,effective and accurate,which can be used for the simultaneous determination of 8 active constituents in Biyan Qingdu granules.
