摘要
目的:观察双重动脉血供(LDABS)对肝脏线粒体能量代谢的影响。方法:108只Sprague-Dawley(SD)大鼠(购自北京维通利华公司)采用完全随机化方法分分3组,即假手术组(SO组)、68%肝切除组(PH组)、68%肝切除结合双重动脉血供组(PH+LDABS组,简称LDABS组)。每组模型术后0、12、24、72、120和168 h各组每个时间点随机取材6只;通过测线粒体膜电位、呼吸链酶浓度及三磷酸腺苷(ATP)酶探索LDABS致线粒体能量代谢的影响,组间比较用单因素方差分析,组间两两比较用LSD检验。结果:线粒体呼吸链酶Ⅰ在LDABS组术后12 h和72 h[(0.48±0.02,0.33±0.02)μmol/(min·mgprot),t=-5.296、-6.399,P值均<0.05],差异有统计学意义;Ca^2+-Mg^2+-ATP酶在LDABS组术后168 h低于PH组[(35.84±5.02)U/g,t=8.857,P<0.01];术后24 h高于SO组[(37.33±5.39)U/g,t=-0.127,P<0.05],差异均有统计学意义;Na^+-K^+-ATP酶在LDABS组术后12 h达到峰值,术后0 h高于PH组[(20.27±0.69)U/g,t=-4.527,P<0.01],差异有统计学意义。结论:LDABS可能通过线粒体呼吸链酶Ⅰ在肝再生能量代谢中发挥重要作用,可能在更早的阶段通过提高能量代谢促使肝细胞再生,且可能使肝再生所需的能量代谢的峰值提前。
Objective Through the establishment of a model,the effect of double arterial blood supply(LDABS)on liver mitochondrial energy metabolism was discussed.Methods 108 SD rats were purchased from Beijing Viton Lihua Experimental Animal Technology Co.LTD.They were randomly divided into three groups:sham operation group(SO group),68%partial hepatectomy group(PH group),68%hepatectomy combined with dual arterial blood supply group(LDABS group).Each group of animal models were taken at 0,12,24,72,120 and 168 hours after operation.The effects of mitochondrial membrane potential,respiratory chain enzyme concentration and ATPase analysis on mitochondrial energy metabolism of LDABS were explored.One way ANOVA was used for comparison between groups,and LSD test was used for pairwise comparison between groups.Results Mitochondrial respiratory chain enzyme I was 12 h and 72 h after LDABS group[(0.48±0.02,0.33±0.02)μmol/(min·mgprot),t=-5.296,-6.399,P<0.05],the difference was statistically significant;Ca^2+-Mg^2+-ATPase was lower than PH at 168 h after LDABS group Group[(35.84±5.02)U/g,t=8.857,P<0.01];24 h after operation was higher than that of SO group[(37.33±5.39)U/g,t=-0.127,P<0.05],the difference was statistically significant;Na^+-K^+-ATPase reached its peak at 12 h after LDABS group,0 h after operation was higher than the PH group[(20.27±0.69)U/g,t=-4.527,P<0.01],the difference was statistically significant.Conclusion LDABS may play an important role in the energy metabolism of liver regeneration through mitochondrial respiratory chain enzyme I.It may promote hepatocyte regeneration by improving energy metabolism at an earlier stage,and may advance the peak of energy metabolism required for liver regeneration.
