螺内酯改善代谢综合征大鼠血管内皮功能的实验研究 被引量:1

摘要 目的观察螺内酯改善代谢综合征大鼠血管内皮功能的作用。方法选取高果糖饮食饲养的16只雄性SD大鼠建立代谢综合征模型,将造模成功后大鼠随机分为代谢综合征组和螺内酯干预组,并选取普通饲料喂养的8只雄性SD大鼠作为正常对照组,采用硝酸还原酶法检测血浆一氧化氮(NO)含量,采用放射免疫法检测血清单核细胞趋化蛋白-1(MCP-1)含量,采用免疫组织化学法检测主动脉一氧化氮合成酶(eNOS)表达。结果高果糖饮食成功诱导高血压、高三酰甘油血症和高血糖等代谢综合征模型,代谢综合征大鼠血浆NO浓度降低(P<0.01),血管内皮eNOS表达同步下调,血清MCP-1浓度升高;采用螺内酯干预后,内皮功能有所改善。结论螺内酯可改善代谢综合征大鼠血管内皮功能,其机制可能与抑制炎症反应有关。 Objective To observe the effect of spironolactone on vascular endothelial function in rats with metabolic syndrome(MS).Methods The MS rats were induced by high-fructose diet,and the modeling rats were randomly divided into model control group and spironolactone intervention group.The level of nitric oxide(NO)was detected by nitrate reductase assay,the expression of monocyte chemotactic protein-1(MCP-1)was detected by radioimmunoassay,and the expressions of nitric oxide synthetase(eNOS)was determined by immunohistochemistry.Results The models of hypertension,hyperglycemia and other metabolic syndrome were induced by high fructose diet successfully.The levels of NO and eNOS in rats with MS were decreased(P<0.01),the expression of MCP-1 was increased.Endothelial function was improved after intervention with spironolactone.Conclusion Spironolactone could improve vascular endothelial function in rats with MS,and the mechanism might be related to the suppression of inflammation.
