宏基因组来源的酯酶酶学性质及对邻苯二甲酸酯的降解

摘要 邻苯二甲酸酯(PAEs)是一类全球性的重要环境污染物。找到广谱高效降解PAEs且具有良好环境适应性的降解酶尤为重要。利用宏基因组学技术,从土壤中克隆出一种新型酯酶基因est924,与已报道的来源于Synechococcus sp.CC9311的alpha/beta-水解酶同源性最高,为62.42%。特异性扩增酯酶基因est924,与质粒载体pET-41a(+)连接,然后将重组质粒转化入大肠杆菌BL21(DE3)中进行异源表达,获得重组酯酶Est924,并研究其酶学性质和对PAEs的降解作用。研究表明该酶是一个具有良好稳定性和环境适应性的耐碱酯酶,并对邻苯二甲酸二甲酯(DMP)、邻苯二甲酸二乙酯(DEP)、邻苯二甲酸二丙酯(DPrP)、邻苯二甲酸二丁酯(DBP)、邻苯二甲酸二戊酯(DPP)以及邻苯二甲酸二己酯(DnHP)等多种PAEs均具有较好的降解活性。因此,酯酶Est924对PAEs污染环境具有巨大的修复潜能。 Phthalate esters(PAEs)are a kind of important global environmental pollutants.It is particularly important to find a broad spectrum of enzymes that degrade PAEs efficiently and have good environmental adaptability.A novel esterase gene est924 was cloned from microorganisms in soil by metagenomic technique,with the highest homology of 62.42%to the reported alpha/beta-hydrolase derived from Synechococcus sp.cc9311.The gene encoding est924 was obtained by PCR amplification,the recombinant plasmid was obtained by ligating the plasmid pET-41a(+)with est924.Then,the recombinant plasmid was transformed into E.coli BL21(DE3)for heterologous expression,and the recombinant esterase Est924 was obtained.The enzymatic properties of Est924 and its efficiency to degrade PAEs were studied.The results showed that Est924 obtained in this study was a new alkali-resistant esterase and had good degradation activities to dimethyl phthalate(DMP),diethyl phthalate(DEP),dipropyl phthalate(DPrP),dibutyl phthalate(DBP),diamyl phthalate(DPP)and dihexyl phthalate(DnHP).Therefore,Est924 has a great potential for PAEs bioremediation.
