摘要
肝素的结构通常以不同的二糖单位表示,了解肝素二糖的组成有助于从微观结构上解析肝素的作用机理。采用肝素酶Ⅰ、Ⅱ、Ⅲ混合,在合适的温度及缓冲液条件下,共同酶解肝素溶液,肝素酶解后得到的二糖及寡糖由于其磺酸化程度的不同因而在阴离子交换柱上保留的程度不同,已达到分离的效果,通过二糖标准品对肝素酶解后的二糖及寡糖进行定位,进而确认肝素酶解后不同糖的构成比例。结果表明,肝素结构测定检测方法具有较好的精密度,在分别改变柱温、体积流量时具有较好的耐用性,该检验方法可用于肝素二糖结构的解析。
The structure of heparin usually is represented by different disaccharide units.It can help learn the mechanism of heparin in the microstructure by analysis of disaccharide structure.Mix the heparin enzyme I,II,III to decompose the heparin solution with an appropriate temperature and buffer solution.The retention degrees of acquired disaccharide and oligosaccharide are different in the anion exchange column because of different sulfonation degrees.So disaccharide and oligosaccharide can be separated.Ensure the location of disaccharide and oligosaccharide by disaccharide standard then calculate the percentage of different disaccharide and oligosaccharide unit after enzymolysis.The result shows that this test method is with qualified precision and robustness when changing column temperature and flow rate.So this method can be used to analyze the disaccharide structure of heparin.
