摘要
为了解猪流行性腹泻病毒(PEDV)的遗传变异情况,本研究对2016—2018年间从河南和山西等地多个规模化猪场随机采集的25份患有腹泻仔猪小肠及内容物提取RNA进行PEDV RT-PCR检测,阳性样品进行S全基因克隆、遗传进化与重组分析。结果显示,25份腹泻仔猪样品中检测出18个PEDV阳性样品,成功克隆18株PEDV毒株S全基因序列,与GenBank中发表的30条PEDV S基因序列进行比较分析,核苷酸同源性均在92.8%以上,存在多处碱基点突变、插入与缺失,且SX-TY1-2017毒株在3291~4132 nt位置处发生重组,可能是由亲本株CH/HNZZ47/2016和HN-KF-2017重组产生的。基于S全基因遗传进化树结果,克隆的18株PEDV属于G2群且位于不同分支,表明PEDV流行株在2016—2018年间出现一定的变异。
In order to understand the genetic variation of the porcine epidemic diarrhea virus(PEDV).Reverse transcription-polymerase chain reaction(RT-PCR)was detected from RNA extracted from the small intestine and contents of 25 piglets with suspected diarrhea randomly collected from several large-scale pig farms in Henan and Shanxi between 2016 and 2018.The S gene was cloned from RNA extracted from the positive PEDV samples,and sequenced and performing for recombinant analysis.The results showed that 18 samples among the 25 diarrhea-affected piglet samples were positive for PEDV detection(72%).The S gene sequence of 18 PEDV epidemic strains was successfully cloned,and the nucleotide homology can be more than 92.8%compared with the 30 PEDV S gene sequences published in GenBank.There were multiple point mutations,base insertions and deletions,and the SX-TY1-2017 strain was recombined at the position of 3291-4132 nt.It may have been formed by parent strains CH/HNZZ47/2016 and HN-KF-2017.The phylogenetic analysis based S gene revealed the 18 epidemic strains belonged to the G2 group and were located in different branched,indicating that PEDV S gene has some variation between 2016 and 2018.
